电针结合康复训练对缺血脑卒中大鼠VEGF与MMP2表达的影响 被引量:4

Effect of electroacupuncture combined with rehabilitation training on the expression of VEGF and MMP2 in the hippocampus of ischemic stroke rat

摘要 目的探讨电针结合康复训练治疗对脑卒中大鼠学习记忆能力及大鼠海马血管内皮生长因子(VEGF)和基质金属蛋白酶-2(MMP2)表达的影响。方法 60只SD大鼠随机分为3组:假手术组、模型组与电针+康复训练组,采用大脑中动脉阻塞法(middle cerebral artery occlusion,MCAO)建立局灶性脑缺血动物模型。采用Morris水迷宫实验评估大鼠的学习、记忆能力,免疫组织化学与Western blot方法检测大鼠海马VEGF与MMP2表达,real time PCR方法检测大鼠海马VEGF与MMP2基因表达水平。结果与模型组相比,电针+康复训练组大鼠在实施治疗后,逃避潜伏期明显缩短(P<0.01)。与假手术组相比,模型组大鼠海马VEGF与MMP2蛋白和mRNA表达水平显著升高(P<0.01),而与模型组相比,电针+康复训练组大鼠海马VEGF与MMP2蛋白和mRNA表达水平显著降低(P<0.01)。结论电针结合康复训练能够改善脑卒中大鼠的学习记忆能力,并能降低大鼠海马VEGF与MMP2的表达。 Objective To investigate the effect of electroacupuncture combined with rehabilitation training on the ability of learning and memory of rats with ischemic stroke and the expression of vascular endothelial growth factor(VEGF) and matrix metalloproteinase 2(MMP2) in the hippocampus. Methods 60 SD rats were randomly divided into 3 groups: sham operation group, model group and electroacupuncture plus rehabilitation training group, and middle cerebral artery occlusion(MCAO) was used to establish focal cerebral ischemia animal model. The ability of learning and memory of rats was evaluated by morris water maze, the expression of MMP2 and VEGF in the hippocampus was detected by immunohistochemistry and Western blot, the expression of VEGF and MMP2 genes in hippocampus was detected by real time PCR. Results Compared with the model group, the escape latency was significantly shortened in electroacupuncture plus rehabilitation training group(P〈0.01). The expression levels of VEGF and MMP2 protein and m RNA in hippocampus were increased significantly in model group than in sham operation(P〈0.01), but decreased significantly in electroacupuncture plus rehabilitation training group than in model group(P〈0.01). Conclusion electroacupuncture combined with rehabilitation training improved the neural function of cerebral apoplexy rats and decreased the expression of VEGF and MMP2 in the hippocampus of rats.
