CHD1L在星形细胞瘤中的表达及其与临床病理因素关系的研究

摘要 目的染色质解旋酶DNA结合蛋白1样基因(CHD1L)是一种新发现位于染色体1q21的致癌基因,在多种恶性肿瘤中都有异常高表达的现象,本文探讨CHD1L在人星形细胞瘤中的表达水平及其与星形细胞瘤患者临床病理因素的关系。方法选取2006-2011年间星形细胞瘤术后石蜡标本120例,组织学分级I-IV级,通过免疫组织化学染色的方法检测CHD1L在星形细胞瘤中的表达水平。制定评分标准,并由三名病理学医师对染色结果进行评分,应用卡方检验统计CHD1L的表达与患者临床病理因素的关系,Kaplan-Meier分析CHD1L的表达与患者预后的关系。结果 CHD1L在星形细胞瘤中异常高表达,其高表达与肿瘤体积、WHO分级及Ki-67指数呈正相关(P<0.05),而与年龄、性别和肿瘤的发生部位没有明显关系。Kaplan-Meier曲线表明CHD1L的高表达与星形细胞瘤患者不良预后正相关。结论 CHD1L可能在星形细胞瘤的发生发展中发挥作用。 Objective The chromodomain helicase/ATPase DNA binding protein 1-like gene(CHD1 L) is a newly discovered gene, located on the chromosome 1 q21. The abnormally high expression CHD1 L was detected in various malignancies. However, the expression and significance of CHD1 L in glioma is still unclear, the purpose of this study is to investigate the expression of CHD1 L and its relationship with clinicopathological features in glioma patients. Methods 120 cases of gliomas were selected from 2006 to 2011 with WHO classification grade Ⅰ-Ⅳ. The expression level of CHD1 L in gliomas was detected by immunohistochemical staining. The relationship between the expression of CHD1 L and clinicopathological features in glioma patients was analyzed by Chi-Squared Test. The relationship between CHD1 L and the prognosis of the patients was tested by Kaplan-Meier analysis. Results CHD1 L was highly expressed in gliomas, and positively correlated with the tumors size, WHO grade and Ki-67 index(P〈0.05), but not correlated with age, gender and tumor location. The Kaplan-Meier curve showed that the high expression of CHD1 L was positively correlated with the poor prognosis of glioma patients. Conclusion CHD1 L might he involved in the development and progression of glioma.
