异丙肾上腺素诱导小鼠心肌肥厚模型的方法优化 被引量:4

The method optimization of cardiac hypertrophy model in mice induced by isoproterenol

摘要 目的对应用异丙肾上腺素(isoproterenol,ISO)诱导小鼠心肌肥厚模型的方法进行进一步的优化。方法小鼠皮下注射剂量为7mg/kg的ISO,每日注射1次,连续注射1周。应用心重体重比,左心室重体重比,心肌细胞横截面积等指标来验证心肌肥厚模型的构建情况。应用切片HE染色方法来观察小鼠心肌细胞的形态学改变。结果实验组与对照组相比,心重体重比,左心室重体重比,心肌细胞横截面积均增加(P<0.05)。与对照组相比,实验组心肌细胞横截面的直径增大,细胞间质增宽,细胞核变形。结论皮下注射ISO 1周,成功构建小鼠心肌肥厚模型,该方法更加简单,经济,科学,快速。为后续实验进行更加深入的研究提供了条件。 Objective To optimize the way of utilizing isoproterenol to induce cardiac hypertrophy model in mice. Methods 7 mg/kg ISO was subcutaneously injected into mice, once a day, for one week. The ratio of heart weight and body weight, the ratio of left ventricle weight and body weight and the cross-sectional area of cardiomyocytes were measured to identify the constructing situation of cardiac hypertrophy. HE staining was used to observe the morphologic changes of cardiomyocytes. Results Compared with the control group, the ratio of heart weight and body weight, the ratio of left ventricle weight and body weight and the cross-sectional area of cardiomyocytes were increased in model group(p〈0.05). Compared with the control group, the cardiomyocytes had a larger cross-sectional area, a wider intercellular matrix and a shape-changed nucleus in model group. Conclusion Subcutaneous injection of ISO induced a mice cardiac hypertrophy model successfully, and was more easy, economic, scientific and quick.
