TRPC6和α-actinin-4在高脂饮食C57BL/6J幼鼠肾小球中的表达及意义

摘要 目的探讨高脂饮食对幼鼠肾小球TRPC6和α-actinin-4蛋白表达的影响及意义。方法 36只3周龄雄性C57BL/6J幼鼠随机分为正常饮食组和高脂饮食组,分别给予标准饲料与高脂饲料喂养12周。通过免疫组织化学和免疫印迹技术检测TRPC6和α-actinin-4蛋白在小鼠肾小球中的表达变化。结果与正常饮食组相比,TRPC6蛋白在高脂饮食组C57BL/6J小鼠肾小球中表达显著升高,而α-actinin-4蛋白的表达却显著降低。结论在高脂饮食诱导的肾小球损伤中,TRPC6蛋白的高表达可能干扰了骨架蛋白α-actinin-4的正常表达,破坏了肾小球结构的稳定性,进而导致肾小球损伤。 Objective To investigate the expression and significance of TRPC6 and α-actinin-4 in glomerulus of high-fat diet C57BL/6J mice. Methods 36 male C57BL/6J mice(3-week old) were randomly assigned into two groups. The standard group was fed with basic diet and high-fat diet group was fed with high-fat diet for 12 weeks. The expressions of TRPC6 and α-actinin-4 were examined by immunohistochemistry and western blotting. ResultsCompared with standard group, expression of TRPC6 of high-fat diet group was increased in glomerulus, and the ecpression of α-actinin-4 was reduced. Conclusion The higher expression of TRPC6 in glomerular injury induced by high-fat diet might interfere with the normal expression of α-actinin-4, influence the stability of glomerulus, and result in glomerulus injury.
