miR-181b靶向NDRG2对胃癌细胞SGC-7901侵袭迁移能力的影响 被引量:4

Expression and effect of miR-181b in gastric carcinoma on the proliferation and invasion ability of gastric cancer cell line SGC-7901

摘要 目的研究miR-181b靶向NDRG2对胃癌细胞SGC-7901侵袭迁移能力的影响。方法运用qPCR法检测miR-181b在胃癌及癌旁组织中的表达,运用Western blot法检测NDRG2在胃癌组织及癌旁正常组织中的表达,用双荧光素酶报告基因系统检测miR-181b对NDRG2转录活性的影响,用Transwell实验检测过表达miR-181b对SGC-7901细胞侵袭迁移能力的影响,用Western blot法检测过表达miR-181b后SGC-7901细胞NDRG2的表达变化。结果与癌旁组织比较,胃癌组织miR-181b的表达明显升高,NDRG2蛋白表达水平明显降低(P<0.01)。双荧光素酶报告基因系统检测结果显示,miR-181b可以直接调控NDRG2的转录活性。过表达miR-181b后,SGC-7901细胞的侵袭和转移能力明显升高,NDRG2的表达水平下调(P<0.01)。结论 miR-181b可靶向NDRG2的表达调控SGC-7901细胞的侵袭迁移能力。 Objective To investigate the effect of miR-181b targeting N-myc downstream regulated gene 2(NDRG2) on the invasion and migration of gastric cancer cell SGC-7901. Methods The expression of miR-181b in gastric cancer and adjacent tissues was detected by qPCR. The expression of NDRG2 in gastric cancer and adjacent tissues was detected by Western blot. The effect of miR-181b on the transcriptional activity of NDRG2 was determined by double luciferase reporter gene system. The transwell test was used to detect the ability of invasion and migration of SGC-7901 cells after overexpression of miR-181b. The expression of NDRG2 in SGC-7901 cells was detected by Western blot after overexpression of miR-181b. Results Compared with the normal tissues, the expression level of miR-181b in gastric cancer tissue was significantly increased, and the expression level of NDRG2 protein was significantly decreased(P〈0.01). miR-181b could directly regulate the transcriptional activity of NDRG2. After overexpression of miR-181b, the invasion and metastasis of SGC-7901 cells increased significantly, and the expression level of NDRG2 was down-regulated(P〈0.01).Conclusion miR-181b could regulate the invasion and migration of SGC-7901 cells by targeting NDRG2.
