IL-6基因启动子区单核苷酸多态对京海黄鸡柔嫩艾美尔球虫抗性指标的影响 被引量:7

The effects of IL-6 gene promoter region single nucleotide polymorphism to Eimeria tenella resistance index in Jinghai yellow chickens

摘要 为探究白介素6(IL-6)基因5′启动子区单核苷酸突变对鸡柔嫩艾美尔球虫抗性指标的影响,以京海黄鸡为试验材料,对IL-6基因5′调控区单核苷酸多态性(SNPs)进行检测,根据感染组与对照组血浆抗性指标酶联免疫吸附试验检测结果,分析SNPs突变形成的基因型与抗性指标的关联。结果表明:感染组与对照组间过氧化氢酶(CAT)、谷胱甘肽过氧化物酶(GSH-Px)活性和丙二醛(MDA)含量差异极显著,超氧化物歧化酶(SOD)活性及IL-2、IL-16、IL-17、γ干扰素(IFN-γ)水平和体增重差异显著;通过京海黄鸡IL-6基因启动子区基因序列与GenBank上的SNPs数据库比对,发现3个新的SNPs(A-663G、C-447G和G-357A),其中A-663G位点只有1种基因型GG,C-447G和G-357A位点均处于Hardy-Weinberg平衡状态且多态性丰富;C-447G突变位点各基因型间比较发现,血浆CAT、SOD、GSH-Px活性及IL-2、IL-16、IL-17水平和体增重差异极显著,MDA含量差异显著;G-357A突变位点各基因型间比较发现,血浆CAT、SOD活性及IL-16水平和体增重差异极显著,GSH-Px活性和IL-17水平差异显著。就主要抗性指标而言,C-447G和G-357A2个突变位点的CC和AA纯合型个体优于其他基因型个体,CC和AA基因型分别为2个突变位点鸡球虫抗性有利基因型,这一结果为京海黄鸡鸡球虫抗性分子标记辅助育种提供了参考。 In order to explore the influence of single nucleotide polymorphisms(SNPs)in 5′promoter region of IL-6 on the resistance of Eimeria tenella,the Jinghai yellow chicken was used as experimental material to detect the SNPs in the 5′promoter region of IL-6 by DNA sequencing.According to the results of the enzyme-linked immunosorbent assay(ELISA)of the plasma resistance markers between the infected group and the control group,the association of genotypes and resistance markers formed by mutations in SNPs was analyzed.The results showed that the difference of CAT,GSHPx and MDA concentrations between the infected group and the control group were extremely significant(P〈0.01).The concentration of SOD,IL-2,IL-16,IL-17,IFN-γand weight gain in the infection group significantly differed from the control group(P〈0.05).Three new SNPs(A-663 G,C-447 Gand G-357 A)were found in the IL-6 promoter region in Jinghai yellow chickens compared with the SNPs database of GenBank,only one genotype GGat the A-663 Glocus was found in Jinghai yellow chickens,and both C-447 Gand G-357 Aloci were in Hardy-Weinberg balance and had rich polymorphic information.The difference of CAT,SOD,GSH-Px,IL-2,IL-16,IL-17 plasmaconcentration and body weight gain between the C-447 G mutation genotypes was highly significant(P〈0.01),the difference of MDA plasma concentration was significant(P〈0.05).The difference of CAT,SOD,IL-16 plasma concentration and body weight gain between the G-357 A mutation genotypes was highly significant(P〈0.01),the difference of GSH-Px and IL-17 plasma concentration was significant(P〈0.05).It could be concluded that the main resistance indices of CCand AA genotypes at the C-447 Gand G-357 Asites were superior to those of other homozygous and heterozygous individuals,and CC,AAgenotype were the dominant genotypes.The results will provide a reference for molecular marker selection breeding of E.tenellaresistance in Jinghai yellow chicken.
