摘要
针对发生在烟草上的马铃薯Y病毒(Potato virus Y,PVY),建立快速、准确且成本低的检测方法。本试验以烟草粗提液和浸提液为反应模板,以硝酸纤维素(nitrocellulose,NC)膜和聚偏二氟乙烯(polyvinylidene fluoride,PVDF)膜为吸附膜,结合RT-PCR检测PVY,通过比较粗提液和浸提液的检出率、NC膜和PVDF膜的检出率,最终建立了以浸提液为反应模板,PVDF膜为吸附膜的浸提液RT-PCR检测方法。该方法检出率与以感病叶片总RNA为模板的检出率一致,耗时更短,成本更低,适用于PVY快速检测。
In order to establish a fast, accurate and low cost method for detecting Potato virus Y(PVY) in tobacco, the experiment was conducted with reaction templates of crude extract and lixivium of tobacco, using adsorption membrane of nitrocellulose(NC) and polyvinylidene fluoride(PVDF) through RT-PCR. Based on comparing of detection rates between the crude extract and lixivium and between NC membrane and PVDF membrane, detection method was established with templates of the lixivium and adsorption membrane of PVDF. And detection rate of this method was in accordance with the method with template of total RNA from the PVY infected leaves, which was less time-consumed and lower cost and suitable for rapid detection for PVY.
