二甲双胍联合利拉鲁肽对多囊卵巢大鼠炎性因子及性激素水平的影响

Effect of metformin combinated with liraglutide on changes of inflammatory factors and hormonal readiness in PCOS rats

目的研究二甲双胍联合利拉鲁肽对经脱氢表雄酮诱导的多囊卵巢(PCOS)大鼠外周血清炎性因子及性激素水平的影响。方法将80只PCOS(经脱氢表雄酮诱导)大鼠随机分为对照组(n=40)和干预组(n=40)。对照组大鼠每天给予二甲双胍(270 mg/kg·d)灌胃,干预组大鼠每天给予二甲双胍(270 mg/kg·d)灌胃+利拉鲁肽(5μg/kg·d)皮下注射,连续治疗28 d。治疗结束后测定两组大鼠外周血清炎性因子[白细胞介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)、C反应蛋白(CRP)]、激素水平[卵泡雌激素(FSH)、黄体生成素(LH)、雌激素(E_2)、睾酮(T)]。结果治疗后两组大鼠外周血清IL-6(23.88±7.32)pg/ml、TNF-α(13.24±2.23)ng/L、CRP(1.10±0.19)mg/L、LH(6.57±1.37)mIU/ml、LH/FSH(1.17±0.28)、E_2(1.11±0.18)pg/ml、T(12.21±1.32)ng/d L均较治疗前有所降低且差异有统计学意义(P<0.05),干预组低于对照组,差异有统计学意义(P<0.05);治疗后两组大鼠FSH(5.63±0.77)mIU/ml较治疗前升高,差异有统计学意义(P<0.05),干预组高于对照组,差异有统计学意义(P<0.05)。结论二甲双胍联合利拉鲁肽能很好的降低PCOS大鼠外周血清炎性因子浓度,改善激素水平,为临床实践提供一定的指导。

Objective To study the effect of metformin combined with liraglutide on changes of inflammatory factors and hormonal readiness in polycystic ovary syndrome（ PCOS） rats induced by dehydroepiandrosterone. Methods Eighty rats with PCOS that was induced by dehydroepiandrosterone were randomly divided into control group（ n = 40） and intervention group（ n = 40）. The control group was given by ig melbine（ 270 mg/kg·d）,and the intervention group was given by ig melbine（ 270 mg/kg·d） ＋ lumbar injection liraglutide（ 5 μg/kg·d）. The treatment lasted 28 days. Before and after treatment,the changes of interleukins-6（ IL-6）,tumour necrosis factor-α（ TNF-α）,C-reactive protein（ CRP）,follicule-stimulating hormone（ FSH）,luteinizing hormone（ LH）,LH/FSH,estrogen（ E2）,and testosterone（ T） were observed. Results After treatment,IL-6 was（ 23. 88 ± 7. 32） pg/ml,TNF-α（ 13. 24 ± 2. 23） ng/L,CRP（ 1. 10 ± 0. 19） mg/L,LH（ 6. 57 ± 1. 37） mIU/ml,LH/FSH（ 1. 17 ± 0. 28）,E2（ 1. 11 ± 0. 18） pg/m L,and T（ 12. 21 ± 1. 32） ng/d L,which were all lower than before treatment in both groups（ P〈0. 05）,while FSH（ 5. 63 ± 0. 77）mIU/ml was higher than before treatment in both groups（ P〈0. 05）. Conclusions Metformin combined with liraglutide can reduce the concentration of inflammatory factors and hormonal readiness,which can facilitate clinical practice.
