运脾和络汤对糖尿病大鼠坐骨神经Drp-1介导细胞凋亡线粒体途径的影响 被引量:9

Effects of Yunpi Heluo Decoction on Drp-1 mediated mitochondrial pathway in apoptosis of sciatic nerve in diabetic rats

摘要 目的:观察运脾和络汤对糖尿病大鼠坐骨神经Drp-1介导细胞凋亡线粒体途径的影响,探讨其改善坐骨神经损伤作用机制。方法:将糖尿病模型大鼠分为模型组、阳性组及低、中、高浓度组,设立正常组,每组8只,以对应药物干预16周,坐骨神经切片HE染色观察神经损伤情况,测各组坐骨神经中Drp-1、Bax、Bcl-2、Caspase-9、Caspase-3 m RNA表达,并测坐骨神经中Drp-1蛋白表达。结果:与模型组比较,高浓度组坐骨神经损伤程度减轻,坐骨神经Drp-1、Bax、Caspase-9、Caspase-3 m RNA表达及Drp-1蛋白表达明显下调(P<0.05),Bcl-2 m RNA表达明显上调(P<0.05)。结论:运脾和络汤可下调糖尿病大鼠坐骨神经Drp-1、Bax、Caspase-9、Caspase-3表达,上调Bcl-2表达,改善高糖引起的周围神经损伤。 Objective: To observe the effects of Yunpi Heluo Decoction on Drp-1 mediated mitochondrial pathway in apoptosis of sciatic nerve in diabetic rats. Methods: DM rats were divided into 5 groups(model group, positive group, and low, middle, high doses of Yunpi Heluo Decoction group). The normal control group was set. The animals were treated with corresponding drugs or distilled water by gavage for 16 weeks. Sciatic nerve injury was observed by HE staining. The expression of m RNA, Drp1, Bax, Bcl-2, Caspase-9, and Caspase-3 in sciatic nerve of each group was detected, and the expression of the Drp1 protein in sciatic nerve was measured. Results: The degree of sciatic nerve injury in the high dose group was less than that in the model group. The expression of Drp1, Bax, Caspase-9, Caspase-3 gene and the expression of the Drp1 protein in the sciatic nerve were obviously downregulated(P〈0.05), and the expression of Bcl-2 gene was obviously up-regulated(P〈0.05). Conclusion: The expression of Drp1, Bax, Caspase-9, and Caspase-3 in the sciatic nerve of diabetic rats can be down-regulated by Yunpi Heluo Decoction, and the expression of Bcl-2 can be up-regulated and the peripheral nerve injury caused by high glucose can be improved.
