额尔敦-乌日勒预处理对大鼠心肌缺血再灌注损伤氧化应激的影响 被引量:15

Effects of pretreatment with eerdun wurili on oxidative stress in myocardial ischemia reperfusion injury in rats

摘要 目的:观察额尔敦-乌日勒预处理对大鼠心肌缺血再灌注损伤(MIRI)氧化应激的影响。方法:将60只雄性SD大鼠随机分为6组:假手术组,模型组,额尔敦-乌日勒低、中、高剂量组以及复方丹参滴丸组。除假手术组外,各组大鼠通过结扎冠状动脉左前降支30min,然后松线再灌注120min的方法制备心肌缺血再灌注模型。再灌注结束后取血,取心脏组织,测定血清中血清肌酸激酶(CK)、乳酸脱氢酶(LDH)及谷草转氨酶(AST)的含量;检测心肌组织中超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)、过氧化氢酶(CAT)活性和总抗氧化能力(T-AOC)水平,检测一氧化氮合酶(NOS)、一氧化氮(NO)及丙二醛(MDA)的含量。结果:与假手术组比较,模型组大鼠血清CK、LDH、AST含量显著升高(P<0.01),心肌组织中SOD、GSH-Px、CAT活性、T-AOC水平及NOS、NO含量显著降低(P<0.01),且MDA含量显著升高(P<0.01)。与模型组比较,额尔敦-乌日勒中、高剂量组及复方丹参滴丸组大鼠血清CK、LDH、AST水平显著降低(P<0.01),心肌组织中SOD、GSH-Px、CAT活性,T-AOC水平及NOS、NO含量显著增加(P<0.01,P<0.05),MDA含量显著降低(P<0.01);额尔敦-乌日勒低剂量组心肌组织中SOD、GSH-Px、CAT活性,T-AOC水平及NOS含量显著增加(P<0.01,P<0.05),MDA含量显著降低(P<0.01)。结论:额尔敦-乌日勒对大鼠心肌缺血再灌注损伤氧化应激具有抑制作用,其机制可能为改善抗氧化酶活性,提高氧自由基清除能力,抑制氧自由基产生,通过提高抗氧化能力对大鼠MIRI起到保护作用。 Objective: To observe the effects of pretreatment with eerdun wurili on oxidative stress in myocardial ischemic reperfusion injury(MIRI) in rats. Methods: Sixty healthy male SD rats were randomly divided into 6 groups: sham operation group, model group, eerdun pill low, medium and high dose groups and compound Danshen Pill group. Each group of rats was prepared by ligating the left anterior descending coronary artery for 30 min, then loosening and reperfusion for 120 min to prepare MIRI model, except for the sham operation group. At the end of reperfusion, heart tissue was taken, and blood samples were taken to determine creatine kinase(CK), lactate dehydrogenase(LDH) and aspartate aminotransferase(AST) levels. Superoxide dismutase(SOD), glutathione peroxidase(GSH-Px), catalase(CAT) activity and the level of total antioxidant capacity(T-AOC) were detected in the heart tissue. Nitric oxide synthase(NOS), nitric oxide(NO) and malondialdehyde(MDA) contents were also detected. Results: Compared with sham operation group, the contents of CK, LDH and AST of rats serum in model group were significantly increased(P〈0.01), SOD, GSH-Px, CAT activity, T-AOC level, the contents of NOS, NO in heart tissue decreased significantly(P〈0.01), and the content of MDA increased(P〈0.01). Compared with model group, in eerdun wurili medium and high dose groups and compound Danshen Pill group, the contents of CK, LDH and AST of rats serum were significantly decreased(P〈0.01), SOD, GSH-Px, CAT activity, T-AOC level, the contents of NOS, NO in heart tissue increased significantly(P〈0.01, P〈0.05), and the content of MDA decreased(P〈0.01); in eerdun wurili low dose group, SOD, GSH-Px, CAT activity, T-AOC level, the content of NOS in heart tissue increased significantly(P〈0.01, P〈0.05), and the content of MDA decreased(P〈0.01). Conclusion: Eerdun wurili can inhibit oxidative stress on MIRI rats. The mechanism may be that Eerdun wurili can improve the activity of antioxidant enzymes, increase the oxygen free radical scavenging ability, inhibit oxygen free radicals, and improve antioxidant capacity through the protective effect on MIRI rat.
