摘要
目的:以大鼠创面组织中基质金属蛋白酶(MMP)-2、-9及碱性成纤维细胞生长因子(b FGF)表达变化为切入点,探讨生肌膏治疗急性创面的疗效和对愈合过程不同阶段的作用及部分作用机制。方法:以急性创伤创面SD大鼠模型为研究对象,采用ELISA法对创面肉芽组织中MMP-2、MMP-9在1、3、6、12d的表达进行检测。采用免疫组化法对创面肉芽组织中b FGF在3、6、12d进行观察。观察创面愈合及愈合率。结果:正常组织肉芽中有少量MMP-2表达,创面肉芽组织中MMP-2表达量,1d时两组MMP-2表达升高,3d时两组表达降低,6d时两组表达升高,12d时两组表达降低。两组间比较,3d时实验组高于对照组(P<0.05);6d时实验组显著高于对照组(P<0.01)。正常组织肉芽中有少量MMP-9表达,创面肉芽组织中MMP-9的表达量,1d时两组MMP-9表达升高,3d时实验组表达升高,对照组下降,6d时两组表达降低,12d时两组表达明显降低。两组间比较,3、6、12d时实验组均显著高于对照组(P<0.01)。正常组织肉芽中几乎无b FGF表达,创面组织中b FGF的表达,3d时两组均可见大量的b FGF表达,之后逐渐减低。两组间比较,3、6d时实验组均显著高于对照组(P<0.01)。两组大鼠创面全部愈合。3d时实验组创面愈合率高于对照组(P<0.05);9d时对照组创面愈合率高于实验组(P<0.05);15d时实验组创面愈合率显著高于对照组(P<0.01)。结论:生肌膏可以用于急性创面,在创面愈合的早期(1-6d)和中后期(12-18d)具有促进作用,其部分机制可能与促进创面愈合过程中b FGF及MMP-2及MMP-9的表达有关。
Objective: To observe the expression of MMP-2, MMP-9 and b FGF in granulation tissue of rat's wound, and to explore the effect of Shengji Ointment on the acute wound healing, the effects of different stages of the healing process and partial mechanism of action. Methods: The model of acute wounds of SD rats were used as the research object, the expression of MMP-2 and MMP-9 in wound granulation tissue were detected by ELISA at 1 st day, 3 rd day, 6 th day, 12 th day, the expression of b FGF in wound granulation tissue was observed by Immunohistochemical at 3 rd day, 6 th day, 12 th day. The wound healing time and wound healing rate were recorded. Results: A small amount of MMP-2 expression was observed in normal skin. In wound granulation tissue, MMP-2 expression increased in two groups at 1 d, decreased at 3 d, increased at 6 d, and decreased at 12 d. Comparison between the two groups, the experimental group was higher than the control group at 3 d(P〈0.05). The experimental group was significantly higher than the control group at 6 d(P〈0.01). A small amount of MMP-9 expression was observed in normal skin, In wound granulation tissue, MMP-9 expression increased in two groups at 1 d, increased in experimental groups at 3 d, decreased in control groups at 3 d, the expression of two groups decreased at 6 d, and decreased significantly at 12 d. Comparison between the two groups, the experimental group was significantly higher than the control group at 3 d, 6 d, 12 d(P〈0.01). Almost no b FGF expression in normal skin. In the granulation tissue, large number of b FGF expression at 3 d, then decreased gradually. Comparison between the two groups, the experimental group was significantly higher than the control group at 3 d, 6 d(P〈0.01). The wounds of the two groups were all healed. Wound healing in experimental group was higher than that of control group at 3 d(P〈0.05); control group was higher than the experimental group at 9 d(P〈0.05); experimental group was significantly higher than the control group at 15 d(P〈0.01). Conclusion: Shengji ointment can be used for acute wounds, and can accelerate wound healing in early(1-6 d) and Mid late(12-18 d). The mechanism may be related to the expression of b FGF and MMP-2, 9 in the process of wound healing.
