藏药七十味珍珠丸对帕金森病模型大鼠多巴胺能神经元的保护作用 被引量:7

摘要 目的:探讨藏药七十味珍珠丸(RNSP)对帕金森病(PD)模型大鼠多巴胺能神经元的保护作用。方法:采用6-羟基多巴胺(6-OHDA)右侧纹状体单点注射法制作PD模型大鼠。将48只雄性成年SD大鼠随机分为6组:PD模型组,美多芭组(50mg/kg),RNSP低、中、高剂量组(90、180、360mg/kg),每组8只,另设正常对照组8只。各实验组灌胃给药,每天1次,连续给药4周,PD模型组及正常对照组大鼠分别给予等量0.9%氯化钠溶液。4周后进行旋转实验,免疫组化法观察大鼠黑质致密部络氨酸羟化酶(TH)阳性神经元的表达,高效液相色谱-电化学检测法检测大鼠纹状体内多巴胺(DA)及高香草酸(HVA)含量的变化。结果:与PD模型组比较,美多芭组及RNSP低、高剂量组大鼠旋转频率显著降低(P<0.05)。与正常对照组比较,PD模型组大鼠损毁侧/损毁对侧TH阳性神经元数目比及纹状体内DA含量显著减少(P<0.01,P<0.05);与PD模型组比较,RNSP高剂量组大鼠损毁侧/损毁对侧TH阳性神经元数目比及DA含量显著增加(P<0.05)。结论:RNSP可改善PD大鼠运动功能,增加黑质TH阳性神经元数目及纹状体内DA的含量,对PD模型大鼠多巴胺能神经元具有保护作用。 Objective: To explore the neuroprotective effects of the Tibetan medicine Ranasampel(RNSP) on dopaminergic neurons in Parkinson's disease(PD) rat model. Methods: PD rats were established by micro-injection of 6-hydroxydopamine once into the right striatum. Forty-eight healthy rats were randomly divided into normal group, model group, madopar group(50 mg/kg), RNSP group(90, 180, 360 mg/kg), with 8 rats in each group. Each group was intragastrically administered once a day for 4 weeks, and the model group and normal group were given distilled water at the same volume, once a day for 4 weeks. Then they were assessed with rotation test. The tyrosine hydroxylase(TH) positive neurons were detected by immunohistochemical method, and the contents of dopamine(DA) and homovanillic acid(HVA) in rat's corpus striatum were assessed by high-performance liquid chromatography with electrochemical detection. Results: Compared with the PD model group, the rotation rates in RNSP high dose group, low dose group and the madopar group were significantly decreased(P〈0.05). Compared with the normal group, the number of the TH-positive neurons and the content of DA were significantly reduced in PD model group(P〈0.01, P〈0.05), compared with the PD group, the number of TH-positive neurons and the content of DA increased significantly in RNSP high dose group(P〈0.05). Conclusion: The RNSP could efficiently improve the behavior changes in PD model rats, promote the TH-positive neurons in substantia nigra and the content of the DA in stratum, so as to protect dopaminergic neurons in PD rats.
