高原甘加藏羊发情周期GnRHR基因在垂体和卵巢表达规律的研究 被引量:4

摘要 探究高原甘加藏羊发情周期促性腺激素释放激素受体(GnRHR)基因在垂体和卵巢组织中的表达规律,选取处于繁殖期健康未孕高原甘加藏羊48只,分为4组,每组12只,分别于发情前期、发情期、发情后期、间情期从甘加藏羊垂体和卵巢组织中提取总RNA,应用荧光定量RT-PCR方法,以GAPDH为内参,研究高原甘加藏羊发情周期垂体和卵巢组织中GnRHR基因表达的差异。结果显示:高原甘加藏羊整个发情周期不同阶段GnRHR mRNA在垂体和卵巢组织中均有表达。在甘加藏羊发情前期、发情期、发情后期、间情期垂体组织中GnRHR mRNA的相对表达量依次为0.8246±0.1739、1.5100±0.4187、0.2923±0.0202、1.0000±0.2100,其中发情期的相对表达量最高,与发情后期差异极显著(P〈0.01),与发情前期、间情期差异显著(P〈0.05);在卵巢组织中,GnRHR mRNA的相对表达量分别为1.0910±0.7665、0.5420±0.0763、1.3799±0.7452、1.0000±1.1221,其中发情后期的相对表达量最高,与发情期差异极显著(P〈0.01),与发情前期、间情期差异显著(P〈0.05)。这一研究结果为在分子水平上研究藏羊生殖活动调控机理提供了科学数据。 To explore the expression patterns of GnRHR gene in pituitaries and ovaries of estrous cycle in Ganjia Tibetan sheep,there were 48 sheep selected and 12 were divided into four groups.The total RNA of pituitaries and ovaries respectively were sampled from 48 healthy,non-pregnant Ganjia Tibetan sheep in different stages of estrous cycle and real-time fluorescence quantitative PCR(real time q RT-PCR)technique was used and GAPDH was used as the reference gene,the purpose is to study the different expression of GnRHR gene in pituitaries and ovaries at different stages of estrous cycle in Ganjia Tibetan sheep.The results showed that the GnRHR mRNA was expressed in the pituitaries and ovaries at different stages of the whole estrus cycle Ganjia Tibetan sheep.In the pituitaries,the relative expression of GnRHR mRNA in proestrus,estrus,metestrus and diestrus were respectively 0.8246±0.1739,1.5100±0.4187,0.2923±0.0202 and 1.0000±0.2100.The GnRHR mRNA relative expression in estrus was the highest and was very significantly different from that in metestrus(P〈0.01),and was different from that in proestrus and diestrus(P〈0.05).In the ovaries,the relative expression of GnRHR mRNA were respectively 1.0910 ±0.7665,0.5420 ±0.0763,1.3799 ±0.7452 and 1.0000 ±1.1221.The GnRHR mRNA relative expression in metestrus was the highest and was very significantly different from that in estrus(P〈0.01),and was different from that in proestrus and diestrus(P〈0.05).The study provided scientific data on the regulation mechanism of the reproductive activities of Ganjia Tibetan sheep at the molecular level.
