小麦胚芽中植物甾醇提取条件优化的研究 被引量:4

摘要 采用超声辅助有机溶剂法提取小麦胚芽中的甾醇,并利用HPLC对其成分进行分析。研究了提取溶剂、提取温度、提取时间和液固比对甾醇提取率的影响。在单因素试验的基础上,采用响应面分析法对提取工艺进行优化。结果表明,超声辅助有机溶剂法提取甾醇的最佳工艺条件为提取温度55.99℃、提取时间1.52 h、液固比16.05∶1(m L/g),在此条件下小麦胚芽的甾醇得率为8.99 mg/g。HPLC分析结果表明,小麦胚芽中的甾醇主要为谷甾醇。 Ultrasound-assisted organic solvent extraction was adopted to extract phytosterol from wheat germ, then the composition was analyzed by HPLC. The effects of extraction solvent, extraction temperature, extraction time and ratio of liquid to solid on the extraction of phytosterol were researched. On the basis of single factor experiment, the extraction process was optimized by response surface method( RSM). The results showed that the optimal conditions were as follows: extraction temperature was 55.99 ℃, extraction time was 1.52 h, ratio of liquid to solid was 16.05 ∶ 1(m L/g). Under these conditions, the extraction rate of phytosterol was 8.99 mg/g. The results of HPLC analysis showed that the predominant phytosterol in wheat germ was sitosterol.
