摘要
基于苦荞(Fagopyrum tataricum)花期转录组数据,采用RT-PCR技术克隆到2个编码DREB类转录因子的基因,命名为Ft DREB1和Ft DREB2。氨基酸多重序列比对表明,其编码蛋白Ft DREB1和Ft DREB2具有与拟南芥DREB相同的保守AP2结构域。进化树分析表明,Ft DREB1和Ft DREB2与抗逆相关DREB转录因子归为A2亚家族。实时荧光定量PCR分析表明,在PEG模拟的干旱胁迫下,Ft DREB1和Ft DREB2表达量有所上升,峰值分别在2 h与12 h,分别为对照组的2.09倍(p〈0.01)和2.83倍(p〈0.01);低温与Na Cl胁迫下,Ft DREB1和Ft DREB2表达量均显著下降,趋势基本一致。本研究推测Ft DREB1和Ft DREB2基因以相似的应答模式参与了苦荞对不同非生物胁迫的应答过程。
Based on the transcriptome data of Fagopyrum tataricum in the flowering phase,2 genes encoding DREB transcription factors,named as Ft DREB1 and Ft DREB2,were cloned by RT-PCR technique.The analysis of amino acid multiple sequence alignment showed that Ft DREB1 and Ft DREB2 encoding proteins had the same conserved AP2 structural domain as Arabidopsis thaliana DREB.The real-time FQ-PCR analysis showed that the expression of Ft DREB1 and Ft DREB2 increased under drought stress simulated by PEG.The peak values of Ft DREB1 and Ft DREB2 expression were at 2 h and 12 h,which were 2.09 times(p〈0.01) and 2.83 times(p〈0.01)of the control group,respectively.The expression of Ft DREB1 and Ft DREB2 decreased significantly under low temperature and Na Cl stress,and their change tendency was basically similar.In this study,it was speculated that Ft DREB1 and Ft DREB2 genes would be involved in the response process of Fagopyrum tataricum to different abiotic stresses in a similar response mode.
