原癌基因MYC在乳腺癌中的表达及与预后关系 被引量:5

摘要 目的探讨乳腺癌患者原癌基因MYC的表达及其临床意义。方法行手术治疗的乳腺癌患者135例,采用实时荧光定量PCR法检测乳腺癌组织中MYC mRNA表达水平,Western blot法检测MYC蛋白表达,免疫组织化学法检测MYC蛋白阳性表达率,分析MYC mRNA及蛋白表达与临床病理特征的关系。结果乳腺癌腋窝淋巴结阳性患者、转移或死亡者组织中MYC mRNA(0.49±0.04、0.50±0.06)及蛋白表达(0.79±0.05、0.82±0.04)和MYC蛋白阳性率(84.3%、95.7%)均高于腋窝淋巴结阴性患者和生存者MYC mRNA水平(0.42±0.11、0.44±0.09)、蛋白表达水平(0.71±0.06、0.72±0.09)及MYC蛋白阳性率(59.5%、63.4%)(P<0.05),三阴性乳腺癌组织中MYC蛋白表达(0.80±0.07)高于Luminal型乳腺癌(0.72±0.05)(P<0.05)。结论 MYC在三阴性乳腺癌中表达增高,MYC高表达患者预后较差。 Objective To explore the expression and clinical significance of MYC in breast cancer.Methods Totally 135 patients with breast cancer undergoing operation were detected the expression of MYC mRNA and protein by real-time PCR and Western blot,and the positive expression of MYC protein was detected by immumohistochemistry to analyze the relationship of MYC mRNA and protein with the clinicopathological characteristics.Results The expressions of MYC mRNA(0.49±0.04,0.50±0.06)and protein(0.79±0.05,0.82±0.04)as well as the positive rate of MYC protein(84.3%,95.7%)in breast cancer patients with axillary lymph node positive and metastasis/death were significantly higher than those in patients with axillary lymph node negative and the survivors(MYC mRNA:0.42±0.11,0.44±0.09,MYC protein:0.71±0.06,0.72±0.09;positive rate of MYC protein:59.5%,63.4%)(P〈0.05).The expression of MYC protein was significantly higher in triple-negative breast cancer tissues(0.80±0.07)than that in luminal breast cancer tissues(0.72±0.05)(P〈0.05).Conclusion MYC is highly expressed in patients with triple-negative breast cancer,and the high expression of MYC indicates a poor prognosis.
