miRNA-145、Ki-67、survivin、Aurora-b在喉鳞状细胞癌中的表达及临床意义 被引量:7

摘要 目的探讨miRNA-145、Ki-67、生存素(survivin)、Aurora-b在喉鳞状细胞癌(LSCC)中的表达及临床意义。方法采用实时荧光定量PCR(RT-PCR)检测85例LSCC组织及85例癌旁正常组织中miRNA-145的表达量,采用免疫组化法检测LSCC组织中Ki-67、survivin、Aurora-b的表达水平,分析miRNA-145、Ki-67、survivin、Aurora-b表达情况与LSCC患者临床特征的关系及四者的相关性。结果 LSCC组织中miRNA-145的表达量明显低于癌旁正常组织(P﹤0.01)。LSCC组织中Ki-67、survivin、Aurora-b的阳性表达率明显高于癌旁正常组织,miRNA-145的阳性表达率明显低于癌旁正常组织,差异均有统计学意义(P﹤0.01)。miRNA-145、Ki-67、survivin、Aurora-b的阳性表达率与LSCC患者的TNM分期和淋巴结转移情况有关(P﹤0.01),而与患者的年龄、性别、肿瘤直径及分化程度无关(P﹥0.05)。LSCC组织中miRNA-145与Ki-67、survivin、Aurora-b的表达均呈负相关(P﹤0.01),Ki-67、survivin及Aurora-b的表达两两之间呈正相关(P﹤0.01)。结论 miRNA-145在LSCC组织中低表达,Ki-67、survivin、Aurora-b在LSCC组织中高表达,miRNA-145、Ki-67、survivin、Aurora-b的阳性表达率与LSCC患者的TNM分期和淋巴结转移情况密切相关,四者联合检测可能成为LSCC诊断的有效指标。 Objective To explore the clinical significance and expression of miRNA-145, Ki-67, survivin and Aurorab in laryngeal squamous cell carcinoma(LSCC). Method The expressions of miRNA-145 in 85 cases of LSCC and corresponding adjacent normal tissues were detected by Real-Time PCR. The expressions of Ki-67, survivin and Aurora-b in LSCC and adjacent normal tissues were detected by immunohistochemical method. The relationships among miRNA-145,Ki-67, survivin, Aurora-b and clinical features, as well as the correlation of the four, were analyzed. Result The expression of miRNA-145 in LSCC was significantly lower than that in adjacent normal tissues(P〈0.01), the positive expression rates of Ki-67, survivin and Aurora-b in LSCC were significantly higher than those in adjacent normal tissues(P〈0.01), respectively; the positive rate of miRNA-145 was significantly lower than that in adjacent normal tissues(P〈0.01).The expressions of miRNA-145, Ki-67, survivin and Aurora-b in LSCC were associated with TNM stage and lymph node metastasis(P〈0.01), but not related to age, sex, tumor diameter and differentiation degree(P〈0.05). At the same time, the expression of miRNA-145 in LSCC was negatively correlated with the expressions of Ki-67, survivin and Aurora-b(P〈0.01). Positive correlation was found between the expression of Ki-67, survivin and Aurora-b in LSCC in pairwise comparison(P〈0.01). Conclusion The low expression of miRNA-145, high expressions of Ki-67, survivin and Aurora-b in LSCC tissue are associated with the TNM stage and lymph node metastasis of LSCC, and the combined result of expressions of the four might become an effective indicator for the diagnosis of LSCC.
