摘要
目的体外研究硝苯地平对辛伐他汀代谢的影响。方法不同浓度的硝苯地平与辛伐他汀在人肝微粒体中共孵育,采用超高效液相串联质谱法测定辛伐他汀的浓度,计算硝苯地平对辛伐他汀抑制的IC50值。结果硝苯地平呈剂量依赖性抑制辛伐他汀的代谢,其IC50值为47μmol·L^(-1)。结论硝苯地平可抑制辛伐他汀的代谢,两者联合用药时可能会产生药物相互作用。
Objective To determine the effect of nifedipine on the metabolism of simvastatin in vitro. Methods Simvastatin was incubated with the different concentrations of nifedipine respectively in human liver microsome. The residual concentration of simvastatin in human liver microsome was determined by UPLC- MS/MS. The IC50 of nifedipine's inhibition on the metabolism of simvastatin was calculated by SPSS. Results Nifedipine inhibited the metabolism of simvastatin concentration-dependently, and its IC50 was 47μmol·L^(-1). Conclusions Nifedipine may inhibit the metabolism of simvastatin. Drug-drug interactions may occur in combination of drug use.
