青钱柳总黄酮对3T3-L1前脂肪细胞胰岛素抵抗作用研究 被引量:5

摘要 目的研究青钱柳总黄酮对3T3-L1前脂肪细胞增殖分化及脂肪细胞胰岛素抵抗作用。方法采用MTT法检测青钱柳总黄酮对3T3-L1前脂肪细胞增殖的影响;油红"O"染色检测青钱柳总黄酮对前脂肪细胞分化的影响;应用地塞米松诱导成胰岛素抵抗模型,采用GOD-POD法检测细胞上清液中葡萄糖含量的变化。结果与溶媒对照组比较,64μg·m L-1青钱柳总黄酮作用48 h对3T3-L1前脂肪细胞的增殖和分化效果最佳(P<0.05或P<0.01);与模型组比较,青钱柳总黄酮能显著增加胰岛素抵抗脂肪细胞葡萄糖消耗(P<0.05)。结论青钱柳总黄酮能促进3T3-L1前脂肪细胞增殖和分化,增加脂肪细胞葡萄糖摄取,改善脂肪细胞胰岛素抵抗。 Objective To determine the influence of total flavonoids from Cyclocarya paliurus on the proliferation and differentiation of 3T3-L1 preadipocytes, and its treatment for insulin resistance. Methods The 3T3-L1 preadipocytes were cultured in vitro, and the cell viability was tested by MTT assay. The lipid accumulation of preadipocytes was measured by Oil Red O staining. The insulin resistant cell model was established with dexamethasone. The cellular glucose uptake was determined with GOD-POD assay. Results Compared with the medium control group, total flavonoids at 64 μg·m L-1 increased the proliferation and differentiation of 3T3-L 1 preadipocytes significantly (P 〈 0.05 or P 〈 0.01) after adding total flavonoids from Cyclocarya paliurus and culturing for 48 h. Compared with the model group, total flavonoids at 64 μg·m L-1 enhanced its glucose uptake significantly (P 〈 0.05). Conclusion Total flavonoids from Cyclocarya paliurus improve the proliferation and differentiation of 3T3-L1 preadipocytes, enhance the cellular glucose uptake and promote the secretion of adiponectin insulin resistant adipocytes.
