补肾健脾活血方对高糖环境下MC3T3-E1细胞增殖及成骨分化影响研究 被引量:3

摘要 目的:探讨补肾健脾活血方对高糖环境下MC3T3-E1细胞的增殖和成骨分化的影响。方法:以MC3T3-E1细胞为研究对象,制备补肾健脾活血方含药血清,实验分为空白对照组、高糖组、中药低剂量组、中药中剂量组、中药高剂量组、二甲双胍组。采用CCK-8法检测补肾健脾活血方对高糖环境下MC3T3-E1成骨细胞增殖的影响,通过碱性磷酸酶(ALP)活性测定和骨钙素(OC)含量测定来观察补肾健脾活血方对高糖下MC3T3-E1细胞分化能力的影响。结果:与高糖组相比,中药组一定程度上能提高高糖(25 mmol/L)条件下MC3T3-E1细胞的增殖和分化能力,以中药中剂量组效果更为显著(P〈0.05或P〈0.01)。高糖条件下的中药高、中剂量组更有利于成骨细胞的增殖和分化,与其他组相比P〈0.05;且中药高、中剂量组的骨钙素含量较其他组显著增高,P〈0.05。结论:补肾健脾活血方能够促进高糖环境下MC3T3-E1成骨细胞增殖及分化功能。 Objective :To discuss the influence of Bushen Jianpi Huoxue Decoction on MC3 T3-E1 cells' proliferation and differentiation in high glucose environment. Methods :With MC3 T3-E1 cells as the research object,the experiment was divided into blank control group,the high glucose group,low dose group in traditional Chinese medicine(TCM),midde dose group in TCM,high dose group in TCM,the metformin group.The CCK-8 method is used to detect the impact of Bushen Jianpi Huoxue Decoction on MC3 T3-E1 cells' proliferation in high glucose environment. The effects of cell differentiation are observed by alkaline phosphatase(ALP)activity determination and the content of osteocalcin(OC). Results :Compared with the high glucose group,Chinese medicine group with a certain extent(25 mmol/L)can improve MC3 T3-E1 cells proliferation and differentiation ability in high glucose environment,especially to midde dose group in TCM effect more significant(P〈0.05 or P〈0.01). High,middle dose groups in TCM were more beneficial to osteoblast proliferation and differentiation than other group(P〈0.05). Conclusion :Bushen Jianpi Huoxue Decoction has positive effect on the proliferation and differentiation of MC3 T3-E1 cells in high glucose environment.
