miR-96在口腔鳞状细胞癌中的表达及作用的相关研究 被引量:1

Expression and role of miR-96 in oral squamous cell carcinoma

摘要 目的:探讨miR-96在人口腔鳞癌(Oral squamous cell carcinoma,OSCC)组织标本和细胞系中的表达差异。方法:采用实时荧光定量PCR(qRT-PCR)检测人OSCC组织标本和细胞系中miR-96的表达情况,并分析miR-96的表达与患者临床病理特征的相关性。体外转染miR-96抑制剂至OSCC细胞Tca8113中,qRT-PCR检测转染后Tca8113细胞中miR-96的表达变化,CCK-8实验和流式细胞术检测转染后Tca8113细胞的增殖和凋亡情况。结果:qRT-PCR结果显示miR-96在人OSCC组织标本和Tca8113细胞中异常高表达(P<0.01),并且miR-96的表达与临床分期和淋巴结转移密切相关(P<0.01)。转染miR-96抑制剂后,Tca8113细胞中miR-96的表达水平明显降低(P<0.01),细胞增殖能力明显下降(P<0.01),细胞凋亡率明显上升(P<0.01)。结论:miR-96在OSCC组织标本和细胞系中异常高表达,下调miR-96的表达能够明显抑制OSCC细胞的增殖,促进其凋亡。 Objective: To explore the difference in miR-96 expression in human oral squamous cell carcinoma( OSCC) tissue specimens and cell line. Methods: miR-96 expression in human OSCC tissue specimens and cell line was detected using real-time fluorescent quantitative PCR( qRT-PCR),and the correlation of miR-96 expression with the clinicopathological features of patients was analyzed. miR-96 inhibitor was transfected in vitro to the human OSCC cell Tca8113,and changes in miR-96 expression in cells after transfection were detected using qRT-PCR. Meanwhile,changes in cell proliferation and apoptosis after transfection were determined using CCK-8 assay and flow cytometry. Results: qRT-PCR results indicated that miR-96 showed aberrantly high expression in human OSCC cell Tca8113,as well as OSCC tissue specimens( P〈0. 01). Further statistical analysis suggested that miR-96 expression in OSCC tissue specimens was closely related to clinical stage and lymph node metastasis( P〈0. 01). Moreover,miR-96 expression level had remarkably reduced( P〈0. 01) in Tca8113 cell after transfection with miR-96 inhibitor,cell proliferation capacity had markedly lowered( P〈0. 01),and cell apoptosis rate had evidently enhanced( P〈0. 01). Conclusion: miR-96 shows aberrantly high expression in OSCC tissue specimens and cell line. Down-regulating miR-96 expression can apparently suppress OSCC cell proliferation,and enhance its apoptosis.
