摘要 为探究杜鹃不同种之间的亲缘关系,从而为杜鹃属植物准确分类提供参考及杂交选育新品种奠定基础。利用ISSR分子标记技术对28个杜鹃品种进行遗传多样性分析。从50条ISSR引物中筛选出12条多样性较为丰富的引物对供试材料进行DNA扩增,共获得132条清晰条带,其中有127个位点具有多样性,多样性比例为96.21%,平均每条引物可以扩增11个片段和10.58个多态性片段。利用NTSYS 2.1软件进行遗传相似系数计算,其值介于0.285 7~0.925 8之间。基于其遗传距离数据进行UPGMA聚类分析,将28个品种分为3个类群,分类与品种形态学如花型、花期等性状相关联且可以反映出品种间的演变。ISSR扩增图谱差异显著,可用于杜鹃属植物的鉴定与区分。 In order to explore the genetic relationship of Rhododendron species, then providing the reference for the accurate classification and laying a foundation for hybrid breeding of Rhododendron. ISSR molecular marker was used to study the genetic relationships of 28 native Rhodohendron. ISSR fingerprinting amplified by 12 ISSR primers revealed a total number of 132 unambiguous bands, of which 127 ones were polymorphic and the polymorphism frequency was 96.21%. The average primer can be expanded by 11 fragments and 10.58 polymorphisms. As analyzed by NTSYS 2.1, the similarity coefficient between varieties ranged from 0.285 7 to 0.925 8. These 28 varieties of Rhodohendron were divided into 3 groups by UPGMA based on Jacard coefficient. Classification and species morphology such as flower- like, flowering and other traits associated with and can reflect the evolution between species. ISSR amplification map is obvious and can be used for the identification and differentiation of Rhododendron.
