摘要
目的:探讨Th1/Th2细胞失衡以及特异性转录因子表达异常在免疫性血小板减少症(ITP)中的临床意义。方法:入组ITP患者40例,应用流式细胞术检测Th1与Th2细胞、RT-PCR技术检测Th1特异性转录因子T-bet mRNA和Th2特异性转录因子GATA-3mRNA表达水平以及AimPlex流式高通量技术检测Th1细胞因子IFN-γ与TNF-α以及Th2细胞因子IL-4与IL-10水平。20例健康者作为正常对照组。结果:与对照组相比较,ITP组Th1/Th2细胞比值以及Th1细胞因子TNF-α与IFN-γ表达水平显著升高(P<0.05),Th2细胞因子IL-4与IL-10表达水平显著降低(P<0.05)。Th1特异性转录因子T-bet mRNA以及Th2特异性转录因子GATA-3mRNA表达水平显著升高(P<0.05)。结论:ITP患者存在Th1/Th2细胞失衡以及特异性转录因子表达异常,反映了ITP患者机体免疫功能紊乱。
Objective:To investigate the clinical significance of imbalance of Th1/Th2 cells and aberrant expressions of specific transcription factors in patients with immune thrombocytopenia(ITP).Method:Forty patients with ITP were enrolled.Th1 and Th2 cells were detected by flow cytometry(FCM).The expressions of Th1-cytokines IFN-γand TNF-αand Th2-cytokines IL-4 and IL-10 were detected by AimPlex multiple immunoassays for flow.The expressions of Th1-specific transcription factor T-bet mRNA and Th2-specific transcription factor GATA-3 mRNA were detected by RT-PCR.Twenty healthy people served as the normal control group.Result:Compared with the control group,the ratio of Th1/Th2 cells and the expression of Th1-cytokines TNF-αand IFN-γin ITP group were significantly increased(P〈0.05),while the levels of Th2-cytokines IL-4 and IL-10 were significantly decreased(P〈0.05).The expressions of Th1-specific transcription factor T-bet mRNA and Th2-specific transcription factor GATA-3 mRNA were significantly increased(P〈0.05).Conclusion:Patients with ITP have the imbalance of Th1/Th2 cells and the abnormal expressions of specific transcription factors,which reflect the immune dysfunctions of patients with ITP.
