溶血卵磷脂在内皮细胞泡沫化模型中的作用探讨

摘要 目的探讨制作内皮细胞脂质化损伤的泡沫化细胞模型的有效方法并观察初次与再次脂质化损伤后细胞活力的改变。方法观察溶血卵磷脂(lysophosphatidylcholine,lpc)的绝对浓度和lpc相对浓度对细胞存活率的影响,lpc相对浓度包括人脐静脉内皮细胞(human umbilical vein endothelial cells,HUVECs)的接种密度和lpc干预体积。采用油红O检测内皮细胞泡沫化结果。结果 lpc干预后,HUVECs的接种密度与细胞存活率呈正相关,lpc浓度和lpc干预体积与细胞存活率呈负相关。再次脂质化组的内皮细胞比初次脂质化组的细胞存活率更高,且差异有统计学意义。结论综合运用lpc的绝对浓度与相对浓度不同调控参数更有助于制作出一个适宜的内皮细胞泡沫化模型。 Objective To explore the effective method of making the cell foaming model after lipid damage in human umbilical vein endothelial cells( HUVECs) and observe the change of cell vitality after first and second lipid damage. Methods The effect on cell survival rate was observed from the absolute and relative concentration of lysophosphatidylcholine( lpc),the relative concentration of lpc included culturing density of HUVECs and treatment volume of lpc. Foaming result of HUVECs was detected by oil red O staining. Results The culturing density of HUVECs was positively correlated with cell survival rate,but the absolute concentration and treatment volume of lpc were negatively correlated with cell survival rate after lpc treatment.The cell survival rate after second lipid damage was higher than that after first lipid damage,which was statistically significant. Conclusions It helps to make a suitable endothelial cell foaming model through comprehensively using the parameters of absolute and relative concentration of lpc.
