摘要 目的探讨在高海拔地区应用血液基因组DNA提取试剂盒的最佳条件。方法应用RelaxGene Blood DNA System血液基因组DNA提取系统,对109份血样(每份血样分为2组,实验组和对照组)提取DNA,实验组采用优化条件提取DNA、对照组采用试剂盒说明书方法提取DNA。结果实验组109份样品中有13份DNA浓度在20-50ng/μL之间,35份在51-80ng/μL之间,61份样品>80ng/μL;DNA浓度比例高于对照组(P<0.05)。实验组109份样品中DNA纯度OD260/OD280比值在≥1.7-<1.9之内的有105份,比值小于1.7的有3份,比值≥1.9的有1份,DNA纯度比例高于对照组(P<0.05)。结论在高海拔地区采用试剂盒提取血液基因组DNA,需要优化提取条件,所得到的DNA浓度和纯度均能满足下游实验要求,且重复性好。 Objective To explore the optimal conditions for extracting the DNA from blood genomic DNA in the plateau area.Methods RelaxGene Blood DNA System,109 plasma samples(each Blood sample was divided into 2 groups,experimental group and control group)to extract DNA,DNA extracted with the use of optimal conditions and controls kit manual method to extract DNA.Results 13 of the 109 samples of the experimental group were between 20 and 50 ng/μL,35 were between 51-80 ng/μL,and 61 samples were〉80 ng/μL.The concentration of DNA was higher than the control group(P〈0.05).The ratio of the DNA purity OD260/OD280 in the experimental group 109 samples was 105 in the sample,and the ratio was less than 1.7.The ratio was less than 1.7,and the ratio was greater than that of the control group(P〈0.05).Conclusion Uses reagent box extraction blood genome team DNA in the high elevation area,needs to optimize the extraction condition,obtains the DNA density and the purity can satisfy the downstream experiment to request,also the duplication is good.
