摘要
目的:局灶节段性肾小球硬化(focal segmental glomerulosclerosis,FSGS)是临床上常见的一种肾脏疾病,最终可进展至终末期肾病(end-stage renal disease,ESRD)。姜黄素是一种多酚天然产物,从姜黄的根茎中分离出来,具有抗炎和抗氧化的作用。本项研究探索姜黄素对阿霉素诱导的FSGS大鼠的肾脏保护作用。方法:使用阿霉素尾静脉注射(10 mg/kg)诱导FSGS大鼠模型,实验组每日予姜黄素(200 mg/kg)治疗,模型组单纯给予生理盐水。6周后处死大鼠,收集血液,尿液和肾脏标本以做进一步的研究。结果:姜黄素能够激活阿霉素诱导的FSGS大鼠Nrf2和Sirt1基因,进而抑制ROS和炎症因子的产生,延缓纤维化进展,改善肾脏的结构和功能病变。结论:姜黄素可以缓解阿霉素诱导的FSGS大鼠病变进展,其可能的作用靶点为Sirt1和Nrf2两个基因,通过激活Sirt1和Nrf2来抑制炎症反应和氧化应激,进而延缓FSGS的病变进展。
Objective: Focal segmental glomerulosclerosis( FSGS) is a common primary glomerular disease,and it ultimately could develop into end-stage renal disease( ESRD). Curcumin is a natural product of polyphenols,isolated from the rhizome of turmeric,with anti-inflammatory and anti-oxidative effects. This study explored the protective effect of curcumin on Adriamycin-Induced FSGS animal model. Methods: Adriamycin nephropathy was induced by Adriamycin( 10 mg/kg body weight,diluted in normal saline) via a tail vein on day 0. The experimental group was treated with curcumin( 200 mg/kg body weight,diluted in normal saline) daily,and the model group was given pure saline alone. Rats were sacrificed after 6 weeks and blood,urine and kidney were collected for further study. Results: Curcumin could activate the Nrf2 and Sirt1 in Adriamycin-Induced FSGS animal model,then inhibit the production of ROS and inflammatory factor,ultimately delay the progress of renal fibrosis and improve the renal structure and functional lesions. Conclusion: Curcumin could relieve the progress of Adriamycin-Induced FSGS animal model. It may through activating Nrf2 and Sirt1 to inhibit inflammatory response and oxidative stress,thus delaying the progression of FSGS.
