电针和/或二甲双胍治疗多囊卵巢综合征模型大鼠的对比研究

Comparison of Electro-acupuncture and/or Metformin in Treatment of Rats with Polycystic Ovary Syndrome

目的：比较电针、二甲双胍、电针配合二甲双胍治疗对多囊卵巢综合征（PCOS）模型大鼠的作用。方法：40只大鼠随机分成正常对照（D）、模型（M）、电针（A）、二甲（B）组、与二甲针（C）等5组,以丙酸睾丸酮合并高脂饲料造模;分别给予电针（A、C组）和/或二甲双胍（B、C组）治疗,检测各项指标。结果：A、B、C组的睾酮（T）、肿瘤坏死因子-α（TNF-α）、空腹胰岛素（FINS）、和胰岛素抵抗指数（HOMA-IR）显著降低;部分大鼠出现规律的动情周期,黄体数目及卵泡内颗粒细胞层数增加,囊性卵泡减少。结论：A、B、C方案对PCOS大鼠均有疗效。电针结合二甲双胍对糖耐量的调节较单纯电针更快显效。

Objective ： To compare the effects of eletro - acupuncture, metformin, and eletro - acupuncture combined with metformin on the polycystic ovary syndrome （PCOS） rats. Methods： Forty SD rats were randomly designated into five groups, i.e. normal （D） group, model （M） group, eletro - acupuncture （A） group, metformin （B） group, and eletro -acupuncture combined with mefformin （C） group. PCOS was induced in 3 -week -old rats with testosterone propio- nate （TP） subcutaneous injection （10 mg/kg per day for consecutive $ weeks） and high -fat diet feeding （HFD, main- tained to the end of the experiment）. Animals in A and C groups were given electro - acupuncture （2 Hz for 30 minutes per day, five days per week for consecutive five weeks） , while animals in B and C groups were given metformin （250 mg/kg per day for consecutive five weeks）. Blood testosterone（T）, TNF - α, fasting insulin （ FINS）, fasting blood glu- cose （ FBG）, HOMA - IR, and oral glucose tolerance test （OGTI＇） were tested or measured before and after treatment. Results： The blood testosterone, TNF -α, FINS, HOMA -IR, and OGTT （30 -, 60 - min） in the A, B and C groups were lower than that in the M group （P 〈 0. 05 ）, while there was no significant difference in the 120 min OGTT between the A group and the M group. Compared with the M group, the A, B and C groups showed growing follicles and some corpora lutea ovaries. And some rats in A, B and C groups showed normal estrum after intervention. Conclusion： Treat- ments of A, B, and C groups lead to declined TNF-α expression, ameliorated insulin sensitivity, controlled body weight, down - regulated testosterone, and partially recovered estrous cycle. Metformin or metformin combined with elec- tro -acupuncture were better than electro -acupuncture in regulating 120 min OGTT.
