基于结肠黏膜JAK2/STAT3通路研究健脾化痰法的作用机制

摘要 目的:通过观察香砂六君丸对脾虚痰浊大鼠结肠黏膜JAK2/STAT3通路的影响,探讨健脾化痰法的作用机制。方法:40只雄性SD大鼠,随机分为对照组、脾虚痰浊组(模型组)、香砂六君丸低剂量治疗组(香低组)、香砂六君丸中剂量治疗组(香中组)和香砂六君丸高剂量治疗组(香高组),每组8只;全自动生化分析仪检测血清总甘油三酯(TG);ELISA法检测血清超氧化物岐化酶(SOD)、丙二醛(MDA)和脂多糖(LPS)水平及谷胱甘肽过氧化物酶(GSH-Px)活性;免疫组化法检测结肠黏膜磷酸化(p)-JAK2、p-STAT3和Bcl-2表达。结果:与对照组比较,模型组SOD水平、GSH-Px活性和Bcl-2表达均下降,MDA、LPS水平和p-JAK2/STAT3表达升高,均有统计学意义;与模型组比较,后3组SOD水平、GSH-Px活性和Bcl-2表达上升,后2组的MDA水平减少,后3组的LPS水平下降,后3组p-JAK2表达减少,均有统计学差异;与香低组比较,后2组MDA浓度下降,均有统计学意义。结论:健脾化痰法能够对抗氧化应激,抑制内毒素过度释放,干预结肠黏膜细胞JAK2/STAT3通路以增强其抗凋亡能力。 Objective: To study the effect of Xiangsha Liujun Pill(XLP) on colonic mucosal JAK2/STAT3 pathway in spleen deficiency and phlegm turbid rats, attempting to explore the action mechanism of the principle of invigorating spleen. Methods: Forty male SD rats were randomly divided into the control group, spleen deficiency and phlegm turbid group (model group), XLP low- dose treating group (XLG), XLP middle -dose treating group (XMG) and XLP high - dose treating group (XHG), 8 rats in each group. Automatic chemistry analyzer was used to measure serum total glyc- eride level. ELISA was employed to detect serum levels of superoxide dismutase (SOD), malondialdehyde (MDA) and lipopolysaccharide (LPS) and glutathion peroxidase (GSH -Px) aetiviability. The immunohistochemistry was used to detect expressions of phosphorylated (p) -JAK2, p- STAT3 and Bcl -2 in colon muscosa. Results: Compared with those in the control group, SOD level, GSH - Px activitablity and Bcl - 2 expression were decreased, while MDA and LPS levels and p -JAK2/STAT3 expressions were increased significantly in the model groups. Compared with those in the model group, SOD level, GHS -Px aetivitability and Bcl -2 expression in later three groups were increased signifi- cantly, while MDA levels in later two groups and LPS levels and p -JAK2 expression in later three groups were lowered obviously. Compared with that in XLG, MDA levels in later two groups were decreased significantly. Conclusion: The principle of invigorating spleen and resolving phlegm can opposite oxidative stress, inhibit endotoxin overrelease and improve the anti - apoptotic ability of the colonal mueosa via interpreting JAK2/STAT3 pathway.
