莲子心HPLC指纹图谱研究及不同产地生物碱含量测定 被引量:10

摘要 目的:建立莲子心(Plumula Nelumbinis)的HPLC指纹图谱分析方法,为其质量控制及药效物质基础的研究提供依据。方法:采用Agilent Extend-C18(4.6×150 mm,5μm)色谱柱,以甲醇-(10 mM磷酸二氢钾水溶液+0.1%三乙胺)为流动相进行梯度洗脱,流速为1.0 mL/min,检测波长为282 nm,建立莲子心HPLC指纹图谱,运用“中药色谱指纹图谱相似度评价软件2004A版”对9批不同产地的药材进行相似度评价;同时对不同产地的9批莲子心中莲心碱、异莲心碱和甲基莲心碱的含量进行了测定。结果:建立的莲子心HPLC指纹图谱的专属性、精密度、重现性和稳定性均良好;不同产地莲子心样品无论在指纹图谱相似性及生物碱含量方面均存在较大差异;结论:本研究建立了能全面反应莲子心化学成分的HPLC指纹图谱,目前以生物碱类化合物为指标成分评价莲子心质量的方法有待于进一步的研究及改进。 Objective : To establish the HPLC fingerprint of Plumula Nelumbinis and provide a theoretical basis for its quality evaluation. Methods : Agilent Extend - Cls (4.6 mm × 150 mm, 5 μm) column was used with methanol - ( 10 mM potassium dihydrogen phosphate aqueous containing 0.1% triethylamine) as the mobile phase in gradient elution. The flow rate was 1.0 mL/min and the detection wavelength was 282 nm. With the State Pharmacopoeia Commission Similari- ty Evaluation Software (2004A) processing and analysis, 9 batches Plumula Nelumbinis were assessed the similarities ac- cording to similarity evaluation for chromatographic fingerprint. And the content of the Liensinine, isoliensinine and nef- erine were determined. Results : The analytical method of HPLC fingerprint was fluent and reliable with fine repeatability. There were significant differences in the similarity of fingerprints and the content of alkaloids in Plumula Nelumbinis from different habitats. Conclusion: The HPLC fingerprint which could reflect the overall chemical constituents of Plumula Nelumbinis was eatablished, and the method of evaluating the quality of Plumula Nelumbinis with the alkaloid compounds as indicators should be further studied and improved.
