摘要
目的:通过观察电针对变应性接触性皮炎模型小鼠患耳与健耳的耳厚度差、耳重量差、病变组织的病理影响以及周围血中IL-6、IL-10和IL-17的改变,揭示针刺治疗变应性接触性皮炎的组织形态学和免疫学的部分机制。方法:将40只昆明种白色雄性小鼠,随机分成空白组、模型组、西药组、针刺组,每组10只。采用二硝基氯苯刺激法,制作变应性接触性皮炎模型小鼠,观察治疗后小鼠患耳与健耳的耳厚度差、耳重量差、病变组织的病理改变以及外周静脉血中IL-6、IL-10和IL-17的变化。结果:模型组与空白组相比,患耳与健耳的耳厚度差、耳重量差明显增加,病变组织的炎性改变明显加重,IL-6、IL-17明显增加(P〈0.05,有统计意义),IL-10明显降低(P〈0.05,有统计意义);西药组、针刺组与模型组相比较,患耳与健耳的耳厚度差、耳重量差明显降低,病变组织的炎性改变明显好转,IL-6、IL-17明显降低(P〈0.05,有统计意义),IL-10明显增加(P〈0.05,有统计意义)。结论:针刺治疗变应性接触性皮炎的作用机制之一,可能是通过调节外周血中的IL-6、IL-10和IL-17的含量,进而调整Th17/Treg细胞的平衡,从而实现了耳厚度差、耳重量差的降低,病变组织炎性的改善,达到了治疗作用。
Objective : By observing the electroacupuncture on ear thickness, weight and pathological changes of allergic contact dermatitis and health mice and the effects on peripheral blood IL- 6, IL- 10 and IL- 17, to introduce the histo- morphology of acupuncture treatment of allergic contact dermatitis and immunological mechanisms. Methods : Forty white male Kunming mice were randomly divided into blank group, model group, western medicine group, and acupuncture group, each group 10. By using two nitrochlorobenzene stimulation, mice model of allergic contact dermatitis was set up. We observed ear thickness, weight and pathological changes and peripheral blood IL - 6, IL - 10 and IL - 17. Results : Compared with the blank group, the thickness and weight of ears were significantly increased, and the inflammatory chan- ges of the lesion tissues were significantly increased, IL- 6 and IL- 10 significantly increased(P 〈 0. 05 ). IL- 10 was significantly lower(P 〈 0.05 ). Compared with the model group, western medicine group, acupuncture group and the model group' s thickness and weight of ear were significantly decreased, and the inflammatory changes of the lesion tis- sues were significantly improved. IL - 6 and IL - 17 were significantly reduced( P 〈 0.05 ). IL - 10 was obviously in- creasing(P 〈0. 05 ). Conclusion: One of the mechanism of acupuncture treatment of allergic contact dermatitis may be by adjusting the peripheral blood IL - 6, IL - 10 and IL - 17, thereby shifting the balance of Thl7/Treg cells so as to realize the ear thickness and weight reduced and the improvement of inflammatory lesions in the organization to achieve the thera- peutic effect.
