摘要
目的探讨接骨木油对急性肝损伤的预防保护作用及作用机制。方法将60只健康雄性昆明小鼠随机分为6组,即空白对照组、肝损伤模型对照组、阳性对照组(丹酚酸A,SAA)、接骨木油低、中、高剂量组。低、中、高剂量组每日灌胃接骨木油量分别为2.0、4.0和12.0 g/kg,阳性对照组每日丹酚酸灌胃量为7.5 g/kg。小鼠自由摄食饮水,连续喂养28 d。末次给药24 h后,除空白对照组外,其他5组小鼠腹腔注射CCl4-花生油溶液20 mg/kg,16 h后取血清和肝脏样本,进行血清天门冬氨酸氨基转移酶(AST)、丙氨酸氨基转移酶(ALT),肝组织丙二醛(MDA)、超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)生化指标测定。结果与模型组相比,接骨木油低、高剂量组小鼠血清GPT活性降低(P<0.05),接骨木油各剂量组血清GOT活性降低(P<0.05),肝脏MDA含量降低、SOD活性及GSH-Px活性显著提高(P<0.05),且对上述指标的改善作用与阳性对照组效果无差异(P>0.05)。结论接骨木油对小鼠急性肝损伤具有一定保护作用,且作用效果与丹酚酸A基本一致。
Objecttive To study the protective effect of Sambucus willamsii Hance fruit oil on acute liver injury in mice. Methods Sixty male mice were randomly divided into six groups: normal control,model,positive control( salvianolic acid A,SAA) and Sambucus Willamsii Hance fruit oil groups( oil-H,oil-M and oil-L). The mice were daily administered Sambucus willamsii Hance fruit oil at the dosage of 2. 0,4. 0 and 12. 0 g/kg for the groups of oil-L,oil-M,oil-H,respectively,while the same volume of 1% CMC was given to normal control groups and model ones for four weeks. After twenty-four hours from the last administration,the mice( except the normal control group) were injected intraperitoneally with CCl4( 20 mg/kg). All the mice were sacrificed sixteen hours later,and serum and liver of the mice were collected for biochemical index determination.Results The result showed that compared with the model group,the serum aspartate aminotransferase( AST) and alanine aminotransferase( ALT) levels in the Sambucus willamsii Hance fruit oil groups decreased significantly( P〈0. 05). MDA levels reduced( P〈0. 05),while SOD and GSH-Px level increased( P〈0. 05). Moreover,the improvement effect of the above indexes was not different from that of the positive control group( P〉0. 05) Conclusion Therefore,moderate intake of Sambucus willamsii Hance fruit oil exhibits a significantly protective effect on acute liver injury in mice.
