不同来源牛血清样品中HoBi样病毒的检测

Detection of the HoBi-like virus in the bovine serum samples of different origins

为了解HoBi样病毒在我国不同来源的牛血清样品中的存在情况，本研究使用荧光RT—PCR方法对530份进境种牛血清、200份河北省奶牛血清和47份商品化胎牛血清进行检测，并对病毒5′UTR序列进行扩增、测序和遗传进化分析。结果表明，从13份南美源胎牛血清中检出4份HoBi样病毒阳性样品，其余血清均为HoBi样病毒阴性；5′UTR序列分析表明，4株病毒与OenBank中登录的HoBi样病毒聚为一支。进一步分析表明，4株病毒核苷酸同源性为96．9％～100．0％，与南美、欧洲源毒株亲缘关系最近，同源性为95．7％～99．6％，而与亚洲源毒株亲缘关系较远，同源性为88．2％～93．0％。本研究结果表明在我国使用的部分南美源商品化胎牛血清中存在HoBi样病毒的污染。

To investigate the presence of the HoBi-like virus in the bovine serum of different origins in China,the real-time RT-PCR specific for HoBi-like virus was applied to detect the virus in the 530 sera samples of the imported breeding cattle,200 sera samples of the breeding cattle ih Hebei, and 47 commercial foetal bovine sera（FBS） samples. The results demonstrated that 4 HoBi-like viruses were detected from 13 FBS originated in South America,and no virus was detected in other samples. For the 4 HoBi-like viruses,the partial 5′UTR was amplified and sequenced. The phylogenetic analysis demonstrated that the 4 viruses were clearly clustered into the HoBi-like virus available in Genbank,and the nueleotide identity was 96.9%-100.0% within the 4 viruses. The further analysis demonstrated that the 4 viruses had a close relationship with the HoBi-like viruses originated from South America and Europe, with the nucleotide identity ranging from 95. 7% to 99.6% ,while differed genetically from the virus strains from Asia,with a nucleotide identity ranging from 88.2% to 93.0%. The results in the study demonstrated the there was contamination of HoBi-like virus in some South America originated commercial FBS products in China.
