摘要
目的:探讨中脑腹侧被盖区多巴胺神经元在全身麻醉中的作用。方法:80只SD大鼠随机分为毁损组与对照组,在双侧中脑腹侧被盖区(VTA)给予毁损组特异性多巴胺神经元毁损药6-羟多巴胺(6-OHDA)以减少多巴胺神经元。给予对照组双侧VTA等体积的生理盐水。术后2周,比较2组大鼠在丙泊酚及异氟醚全麻状态下翻正反射消失时间(LORR)、翻正反射恢复时间(RORR)及睡眠持续时间。结果:丙泊酚麻醉下,毁损组与对照组比较,LORR时间显著缩短,RORR时间及睡眠持续时间显著延长(P<0.05)。异氟醚麻醉下,毁损组与对照组比较,RORR时间及睡眠持续时间显著延长(P<0.05),LORR时间无明显变化(P>0.05)。结论:在不同药物全身麻醉状态下,中脑腹侧被盖区多巴胺神经元发挥的作用亦不尽相同。
Objective: To determine the role of dopamine neurons in the mesencephalic ventral tegmental area in general anesthesia and influence on sleep duration time. Methods: A total of 80 SD rats were selected. They were divided into the lesion group and the control group by stratified randomized grouping. In the bilateral ventral tegmental area,the dopamine neuron destruction drug 6-hydroxydopamine( 6-OHDA) was used to reduce dopamine neurons in the lesion group. The control group was given the same volume of normal saline in the bilateral mesencephalic ventral tegmental area. 2 weeks after operation,the time of loss of righting reflex( LORR),the time of resumption of righting reflex( RORR) and the sustained period of sleep were compared between the 2 groups of SD rats in the total anaesthesia of propofol and isoflurane. Results: Under propofol anesthesia,the time of LORR of SD rats in the lesion group was significantly shorter than that in the control group,the time of RORR and sleep duration time were significantly longer than that in the control group,and the difference was statistically significant( P〈0. 05). Under the anaesthesia of isoflurane,the time of RORR and sleep duration time of SD rats in the lesion group were significantly longer than that of the control group( P〈0. 05).There was no significant difference in the time of LORR between the lesion group and the control group( P〉0. 05). Conclusion: The role of dopamine neurons in the ventral tegmental area of the mesencephalon is different in the state of general anesthesia in different drugs.
