摘要
将采自武陵山片区的湖南龙山、凤凰、花垣、吉首,以及重庆彭水、武隆等6个县市的烟秆,用95%乙醇室温浸泡的方法提取制备烟碱粗提物,并通过高效液相色谱法检测粗提物中烟碱的含量,以期探究不同地域的烟秆中烟碱含量随海拔高度、纬度不同而变化的规律.采用的色谱柱为Gemini 5u C18柱(250 mm×4.6 mm,5μm),以流动相水(含0.02 mol/L磷酸氢二钠缓冲液,溶剂A,60%)∶甲醇(溶剂B,40%)洗脱;流速1.0 m L/min,检测波长259 nm,柱温30℃,进样量20μL.结果表明,不同地域的烟秆中烟碱含量与海拔高度呈正相关,而与纬度没有明显相关性.实验结果可以为烤烟种植业的废弃物——烟秆的资源化利用提供参考.
The crude extract of nicotine was extracted with 95% ethanol from tobacco stems collected from six counties/city located in Wuling mountain area, i. e. , Longshan County, Fenghuang County, Huayuan County, Jishou City in Hunan Province, Wulong County and Pengshui County in Chongqing City. Nicotine in crude extracts was quantitatively determined by high performance liquid chromatography(HPLC). The purpose of this work was to investigate the relationship between the nicotine content in tobacco stems from different counties/city with the altitude and the latitude where the tobacco stem samples were collected. Chromatography was performed on a reversed-phase Gemini 5u C18 column of 250 × 4. 6 mm i. d. dimensions and 5 μm particle size. The mobile phase consisted of 0. 02 mol/L phosphate buffer in water( solvent A,60% ) and methanol( solvent B, 40% ) with isocratic elution. The flow rate was 1.0 mL/min. The column temperature was set at 30 ℃ and the injection volume was 25 μL. Detection was performed at 259 nm. The resuhs indicated that the contents of nicotine in tobacco stems were positively correlated with the altitudes where the tobacco samples were collected, while there was no significant correlation with their latitudes. This experiment provides a reference for the resource utilization of tobacco stem in Wuling mountain area.
