一种野生哺乳动物肌肉样品的DNA分子鉴定

摘要 测定了1例来自江西井冈山地区的哺乳动物肌肉样品的线粒体细胞色素b(Cytb)基因序列。利用NCBI数据库中的在线Blast功能对所测序列(K001)进行序列一致性搜索,确定为鹿科动物;甄选并下载了鹿科类群中17条及麝科中1条林麝(Moschus berezovskii)的Cytb基因序列,结合本次测定的Cytb基因序列,构建了NJ和ML树;在系统树的基础上,计算了K001与其他部分类群的遗传距离。综合序列一致性、系统树、遗传距离以及动物地理分布的结果,确认本次测定的哺乳动物样品是水鹿(Cervus unicolor),系国家Ⅱ级保护野生动物。鉴定为林业案件的审理提供了科学依据。 Mitochondrial cytochrome b(Cytb) gene sequence was determined from a mammal muscle tissue sample, which was accepted from Jinggangshan district, Jiangxi. The ident search was performed based on Cytb sequence of K001 using online Blast in NCBI database, and it was identified as Cervidae species. Combined with the sequence determined in this study, seventeen Cytb sequences of Cervidae species and one sequence of Moschidae species Moschus berezovskii were screened and downloaded to construct NJ and ML trees. Genetic distances were calculated among some taxa group.Combination of geographical distribution, sequence ident, phylogenetic trees and genetic distance, and the species was identified as Cervus unicolor, which is listed as grade II of national protected animal. The identification will provide scientific reference in solving the trial of forestry cases.
