CD98在结直肠癌组织表达及其与CDl47的关系

Expression of CD98 in colorectal cancer and its relationship with CD147

目的探讨CD98在人结直肠癌组织中的表达、临床意义及其与CDl47的相互关系。方法采用免疫组织化学EnVision两步法检测70例结直肠癌和及其癌旁组织中CD98蛋白的表达。设计和合成针对人CD98基因的特异性小干扰RNA（siRNA），脂质体瞬时转染入HCTll6细胞中，用Westernblot技术检测CD98和CDl47蛋白表达水平。结果CD98在结直肠癌和癌旁组织的阳性率分别为57．1％（40／70）和40．0％（28／70），差异有统计学意义（P=0．042）。结直肠癌中CD98的表达与肿瘤分期（P=0．017）和淋巴结转移（P=0．017）明显相关，而与性别（P=0．125）、年龄（P=0．935）、肿瘤大小（P=0．464）、分化程度（P=0．995）无明显相关。结直肠癌中CD98和CDl47的表达呈正相关（r=0．471，P=0．001）。CD98siRNA能够显著下调HCTll6细胞中CD98和CDl47蛋白表达。结论CD98的表达与大肠癌侵袭转移密切相关，CD98可能通过上调CDl47的表达来调控结直肠癌的侵袭转移。

Objective To investigate the protein exp peopletracellular metalloproteinase inducer （ CD147 ） in human colorectal cancer tissues. Methods EnVision immunohistochemical method was used to detect the expression of CD98 protein in 70 cases of colorectal cancer and their adjacent tissues. Western blotting was used to detect the expression of CD98 and CD147 protein in HCT116 cells after transfeetion with CD98 small interfering RNA （siRNA） and negative control. Results The positive rate of CD98 in colorectal cancer and the adjacent tissues was 57.1% （40/70） and 40.0% （28/70） , which was statisti- cally significant （P = 0. 042 ）. The expression of CD98 in colorectal cancer was closely correlated with tumor stage （P =0. 017） and lymph node metastasis （P =0. 017） , but not with gender （P =0. 125）, age （P =0. 935）, distant metastasis （P =0. 464）, tumor differentiation （P=0. 995）. There was a significant positive correlation between the expression of CD98 and CD147 in colorectal cancer （r = 0. 471, P = 0. 001 ）. The expression of CD98 and CD147 protein was obviously decreased in HCT116 cells after trans- fection with CD98 siRNA, compared with negative control. Conclusion The expression of CD98 is closely related to the invasion and metastasis of colorectal cancer. CD98 may regulate the invasion and metastasis of colorectal cancer by up - regulating the expression of CD147.
