摘要
目的:研究上中下通用痛风汤对尿酸钠诱导大鼠急性痛风关节炎(GA)滑膜组织TOLL样受体(TLR)/髓样分化因子88(MYD88)信号通路相关蛋白表达的影响,揭示上中下通用痛风汤治疗痛风关节炎的作用机制。方法:取60只SD雄性大鼠,按随机数字表法分成对照组、模型组、吲哚美辛组、上中下通用痛风汤高、中、低剂量组6组各10只。除对照组外,其余5组采用尿酸钠晶体溶液踝关节注射建立急性痛风关节炎模型,以上中下通用痛风汤与吲哚美辛对比治疗,观察关节肿胀度、关节滑膜病理变化和关节滑膜TLR/MYD88蛋白表达。结果:上中下通用痛风汤高、中、低剂量在致炎后各时间点均能不同程度地减轻关节肿胀程度,可改善急性痛风关节炎大鼠模型踝关节组织的炎细胞浸润、滑膜组织破坏、血管扩张及充血,滑膜组织中TLR4、MYD88蛋白表达下降,与模型对照组比较差异有统计学意义。结论:上中下通用痛风汤对尿酸钠所致急性痛风关节炎症有较好的抗炎镇痛作用,其作用机制可能与降低炎症信号通路TLR4、MYD88的表达有关。
Objective: To study on the Shangzhongxia Tongfeng Decoction on uric acid sodium induced acute gouty arthritis( GA) in the synovial tissue of TOLL like receptor( TLR)/myeloid differentiation factor 88( MYD88) signal pathway related protein expression,reveal mechanism on Shangzhongxia Tongfeng Decoction in the treatment of gouty arthritis. Methods: 60 male SD rats were randomly divided into 6 groups: control group,model group,indomethacin group,middle and lower Shangzhongxia Tongfeng Decoction group,10 rats in each group. In addition to the control group,the remaining 5 groups were injected with sodium urate crystal solution,ankle joint injection,to establish an acute gouty arthritis model. Contrast treatment of the Shangzhongxia Tongfeng Decoction and indomethacin,observe the swelling degree of joint,the pathological changes of synovial joint and the expression of TLR/MYD88 protein in synovial joint. The results on the Shangzhongxia Tongfeng decootion high,low dose after inflammation at each time point are different degrees of reduced joint swelling degree; can improve the acute gouty arthritis rat model of ankle joint tissue inflammatory cell infiltration,synovial tissue damage,vascular dilatation and congestion; decreased the expression of TLR4 and MYD88 protein in synovial tissue compared with the model group with significant difference. Conclusion: Shangzhongxia Tongfeng decoction has a good anti-inflammatory and analgesic effect on acute gouty arthritis induced by sodium urate,and its mechanism may be related to the reduction of inflammatory signaling pathway TLR4 and MYD88 expression.
