自然衰老小鼠尿样代谢组学研究及FTZ的干预作用

摘要 目的:自然衰老小鼠尿液的内源性代谢变化及贞术调脂方(FTZ)的抗衰老作用机制。方法:将C57BL/6J雌性小鼠分为3月龄对照组、24月龄模型组、24月龄FTZ给药组,收集小鼠的尿液,采用UPLC-MS/MS进行质谱检测,利用主成分分析(Principle Component Analysis,PCA),正交最小二乘法判别分析法(Orthogonal partial least squares discriminant analysis,OPLS-DA)分析衰老小鼠尿液代谢物的变化,筛选潜在标志物及FTZ干预的药效特征及可能作用机制。结果:与对照组相比,模型组小鼠尿液内源性代谢物发生变化的有13种,主要涉及与氨基酸代谢、脂肪酸代谢、烟酸和烟酰胺代谢、能量代谢等,而FTZ给药后上述代谢通路得到显著改善。结论:FTZ具有抗衰老作用,其机制可能与氨基酸代谢、烟酸和烟酰胺代谢、能量代谢等有关,为拓展FTZ的临床应用提供依据。 Objective: Study on the endogenous metabolism of urine in natural aging mice by LC-MS and the efficacy of FTZ. Method: C57 BL/6 J female mice were divided into 3 month old control groups, 24 month old model groups and 24 month old FTZ drug groups. The urine of mice was collected, UPLC-MS/MS was used for mass spectrometry, and Principle Component Analysis Orthogonal partial least squares discriminant analysis was used to analyze the urine metabolic markers of aging mice and the effect characteristics of FTZ intervention. Results: Compared with the control group, there were 13 kinds of endogenous changes in the urine of mice, mainly related to amino acid metabolism, nicotinic acid, nicotinamide metabolism, energy metabolism, and so on, and FTZ was significantly improved. Conclusion: FTZ has antiaging effect, and its mechanism may be related to amino acid metabolism, nicotinic acid, nicotinamide metabolism, energy metabolism and so on. It provides a basis for the development of the clinical application of FTZ.
