人参皂苷Rg_5对胃癌BGC-823细胞增殖、凋亡及凋亡相关因子的影响 被引量:11

摘要 目的观察人参皂苷Rg_5对胃癌BGC-823细胞增殖、凋亡及凋亡相关因子Bcl-2相关X蛋白(BAX)、B细胞淋巴瘤-2(Bcl-2)表达的影响,探讨人参皂苷Rg_5抑制胃癌生长转移的可能作用机制。方法采用CCK-8法检测人参皂苷Rg_5对BGC-823细胞24、48 h的细胞增殖抑制作用,流式细胞术检测高、低浓度人参皂苷Rg_5对BGC-823细胞凋亡的影响,Western blot法检测凋亡相关因子Bax、Bcl-2蛋白的表达。结果与空白组比较,人参皂苷Rg_550、100、200μmol/L 24 h存活率降低(P<0.05),人参皂苷Rg_512.5、25、50、100、200μmol/L 48 h存活率降低(P<0.05);与空白组比较,低浓度人参皂苷Rg_5凋亡率升高(P>0.05),高浓度人参皂苷Rg_5凋亡率升高(P<0.05);与空白组比较,高低浓度人参皂苷Rg_5组Bax蛋白表达均升高(P<0.05),高浓度人参皂苷Rg_5组Bcl-2蛋白表达降低(P<0.05)。结论人参皂苷Rg_5可有效抑制胃癌BGC-823细胞的增殖,促进BGC-823细胞凋亡,且其促进BGC-823细胞凋亡的机制可能与其降低线粒体途径相关的细胞凋亡因子Bcl-2蛋白的表达,升高Bax蛋白的表达相关。 Objective: To study the effect of ginsenoside Rg5 on the proliferation,apoptosis and the expression of Bax and Bcl-2 of BGC-823 cell in gastric cancer,and to investigate the possible mechanism of ginsenoside Rg5 in inhibiting the growth and metastasis of gastric cancer. Methods: CCK-8 method was used to detect the inhibitory effect of ginsenoside Rg5 on the proliferation of BGC-823 cells. Flow cytometry was used to detect the effect of ginsenoside Rg5 on BGC-823 cell apoptosis. Western blot was used to detect the expression of Bax and Bcl-2 protein. Results: Cell proliferation: compared with the blank group,the 24 h survival rates of ginsenoside Rg5 at 50,100 and 200 μmol/L were decreased(P〈0.05);the 48 h survival rates of ginsenoside Rg5 at12.5,25,50,100 and 200 μmol/L were decreased(P〈0.05). Apoptosis: compared with the blank group,the apoptotic rate of low concentration ginsenoside Rg5 was increased(P〈0.05) and the apoptosis rate of high concentration ginsenoside Rg5 was increased(P〈0.05). Western blot results showed that compared with the blank group,the expression of Bax protein in high and low concentrations of ginsenoside Rg5 group were significantly increased(P〈0.05),while the expression of Bcl-2 protein in high concentration of ginsenoside Rg5 group was decreased(P〈0.05). Conclusion: Ginsenoside Rg5 can effectively inhibit the proliferation of BGC-823 cells in gastric cancer and promote the apoptosis of BGC-823 cells,and its mechanism of promoting BGC-823 cell apoptosis may be related to decreasing the expression of Bcl-2 protein and increasing the expression of Bax protein.
