摘要
目的观察消栓饮对创伤性深静脉血栓兔NF-κB的影响并探讨其作用机理。方法选取45只新西兰大白兔,27只采用股静脉钳夹法建造血栓模型后分为消栓饮组(A组)、低分子肝素钠组(B组)、模型组(C组),余下18只均分为假手术组(D组)及空白组(E组),采用ELISA、PCR法检测1、3、7日兔NF-κB含量。结果与假手术组比较,消栓饮组、低分子肝素钠组、模型组的血清NF-κB、静脉壁NF-κB p65 m RNA显著减低,而IKBa m RNA增加(P<0.01);与模型组相比,消栓饮组、低分子肝素钠组血清NF-κB、静脉壁NF-κB p65 m RNA减低,而IKBa m RNA增加(P<0.01)。结论消栓饮可以有效调控血栓形成后NF-κB信号通路,减轻血管壁炎性损伤,提示调控NF-KB信号通路是消栓饮治疗深静脉血栓的机制之一。
Objective: To explore the clinical effect and mechanism of Xiaoshuan Decoction by intervening the changes of NF-κB in rabbits with traumatic deep venous thrombosis. Methods: 45 New Zealand rabbits were randomly selected. A thrombus model was constructed in 27 rabbits with femoral vein clamp method,and they were divided into Xiaoshuan Decoction group(A),low molecular weight heparin group(B) and the model group(C). The remaining 18 were divided into the sham operation group(D) and the blank group(E). ELISA and PCR were used to detect the content of NF-κB on 1 st day,3 rd day and 7 th day. Results: Compared with group D,NFKB and NF-κB p65 m RNA dropped significantly in group A,B and C,and IKBa m RNA increased;the difference was statistically significant(P〈0.01). Compared with group C,NF-κB and NF-κB p65 m RNA dropped significantly in group A and B,and IKBa m RNA increased;the difference was statistically significant(P〈0.01). Conclusion: XSD can effectively reduce the inflammatory injury of blood vessel walls by regulating NF-κB signaling pathways after the thrombus formation in the blood. Regulation of NF-κB signaling pathway is one of the mechanisms of Xiaoshuan Decoction therapy on deep vein thrombosis.
