摘要
目的观察针灸对小鼠过敏性接触性皮炎(ACD)的影响,并探讨其潜在的分子作用机制。方法将小鼠分为正常对照组、模型组、非经络针刺组、经络针灸组、咪康唑组、经络针灸+咪康唑组,每组12只。采用酶联免疫吸附实验(ELISA)检测血清中白细胞介素-2(IL-2)、白细胞介素-4(IL-4)、白细胞介素-7(IL-7)、肿瘤坏死因子-α(TNF-α)的水平;检测各组小鼠背部皮肤肿胀程度以及脏器指数;HE染色检测皮肤组织病理学变化;蛋白免疫印迹(WB)检测皮炎组织中Toll样受体4(TLR4)、核因子κB(NF-κB)的表达水平。结果 HE染色结果显示,正常对照组皮肤结构没有发生明显的变化;模型组皮肤组织间隙水肿,且炎性细胞浸润;与模型组相比,非经络针刺组和咪康唑组皮肤组织没有明显变化,经络针灸组和经络针灸+咪康唑组皮肤组织水肿和炎性细胞浸润情况减轻;经络针灸组血清中IL-2、IL-4、IL-7、TNF-α的含量,皮肤肿胀程度、胸腺指数和脾脏指数,以及皮肤中TLR4、NF-κB表达水平显著低于模型组与非经络针刺组(P<0.05);经络针灸+咪康唑组血清中IL-2、IL-4、IL-7、TNF-α的含量,皮肤肿胀程度、胸腺指数和脾脏指数,以及TLR4、NF-κB表达水平显著低于模型组和咪康唑组(P<0.05);经络针灸+咪康唑组血清中IL-2、IL-4、IL-7、TNF-α的含量,皮肤肿胀程度、胸腺指数和脾脏指数,以及TLR4、NF-κB表达水平与经络针灸组相比,差异无统计学意义(P>0.05)。结论针灸对ACD小鼠具有改善作用,这一作用可能是通过调控TLR4/NF-k B信号通路的活性实现的,为ACD的临床治疗提供一定的依据。
Objective: To explore the effect of acupuncture and moxibustion on allergic contact dermatitis(ACD) in mice and its potential molecular mechanism. Methods: The mice were divided into the normal control group,the model group,non Jingluo acupuncture group,Jingluo acupuncture group,Miconazole group,Jingluo acupuncture plus Miconazole group,12 mice in each group. The levels of serum IL-2,IL-4,IL-7 and TNF-αwere detected by enzyme linked immunosorbent assay(ELISA);the swelling degree of the skin of back and the organ index of the mice were measured;the histopathological changes of skin were detected by HE staining;the expression levels of TLR4 and NF-κB in dermatitis tissue was detected by protein immunoblotting(WB). Results:The results of HE staining showed that there was no obvious change in the skin structure in the normal control group;the skin tissue space of the model group was edema,and inflammatory cell got infiltration;compared with the model group,the skin tissue of the non Jingluo acupuncture group and Miconazole group did not change significantly,the skin tissue edema and inflammatory cell infiltration in the Jingluo acupuncture group and Jingluo acupuncture plus Miconazole group were lighten;the contents of serum IL-2,IL-4,IL-7 and TNF-α,the skin swelling degree,thymus index and spleen index,and the expression levels of TLR4 and NF-κB in the Jingluo acupuncture group were significantly lower than those in the model group and the non Jingluo acupuncture group(P〈0.05);the contents of serum IL-2,IL-4,IL-7 and TNF-α,the skin swelling degree,thymus index and spleen index,and the expression levels of TLR4 and NF-κB in the Jingluo acupuncture plus Miconazole group were significantly lower than those in the model group and the Miconazole group(P〈0.05);the contents of serum IL-2,IL-4,IL-7 and TNF-α,the skin swelling degree,thymus index and spleen index,and the expression levels of TLR4 and NF-κB in the Jingluo acupuncture plus Miconazole group were not significantly different from those in Jingluo acupuncture group(P〈0.05). Conclusion: Acupuncture and moxibustion have an improved effect on ACD mice,which may be achieved by regulating the activity of the TLR4/NF-κB signaling pathway,providing a certain basis for the clinical treatment of ACD.
