摘要
目的探讨松龄血脉康对过氧化氢(H2O2)致人脐静脉内皮细胞(HUVECs)氧化损伤的保护作用和物质基础。方法以500μmol·L-1H2O2溶液建立HUVECs氧化损伤模型,采用CCK-8法检测提取物及含药血清促模型细胞的生长作用;用超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)和丙二醛(MDA)试剂盒检测各组含药血清中SOD、GSH-Px、MDA的表达;采用HPLC法测定含药血清中的成分。结果总黄酮、原花青素及10%松龄血脉康含药血清对模型细胞具有显著的保护作用。与空白组比较,松龄血脉康含药血清中SOD、GSH-Px的表达显著增加,MDA的表达显著降低。松龄血脉康中有7个主要成分入血,其中有4个成分的含量增加;总黄酮与原花青素有3个成分入血,各有1个成分在成品的代谢中受到影响。结论松龄血脉康的7个入血成分可能是其保护血管内皮细胞免受氧化损伤的物质基础。
OBJECTIVE To investigate the protective effects and material base of Sougling Xuemaikang capsule (SXC) on the oxida- tive injury induced by H202 of human umbilical vein endothelial cells(HUVECs). METHODS CCK -8 kit was used to measure cell viability. The activities of SOD, GSH - lax and the content of MDA in the serum were observed by biochemical assay. The HPLC charac- teristic spectrum of medicinal serum of SXC, total flavone (TF) and procyanidine (PC) was determined. RESULTS The SXC, TF and PC medicinal serum accelerated the rate of growth of HUVECs. The activities of SOD and GSH - Px were higher and the contents of MDA were lower in SXC medicinal serum than blank serum. Compared with the SXC characteristic spectrum,there were 7 constituents migrating to blood in SXC medicinal serum ,4 of which were increased. Moreover, there were 3 constituents migrating to blood in TF and PC medicinal serum, respectively, referring to the corresponding compound spectrum. Seven constituents migrating to blood may be the material base of SXC for the improved anti - oxidative ability in HUVECs model. CONCLUSION These results demonstrate that SXC,TF and PC medicinal serum have powerful effects in protecting HUVECs from injury and suggest that protection occurs via a mechanism involving the protection from H202 -mediated oxidative stress.
