黄芩配方颗粒与标准汤剂体外抗炎活性的比较 被引量:15

Comparison of the anti-inflammatory activity of Huangqin dispensing granules and Huangqin standard decoction

摘要 目的:研究黄芩配方颗粒与黄芩标准汤剂的体外抗炎活性,评价两者的药效等效性。方法:在HPLC法对黄芩配方颗粒与黄芩标准汤剂中黄芩苷、汉黄芩苷和黄芩素的含量进行表征的基础上,采用脂多糖诱导小鼠巨噬细胞RAW264.7细胞建立炎症模型,以WST-1法和Griess法分别考察了10批次黄芩配方颗粒及黄芩标准汤剂的细胞毒性及对NO生成量的影响,然后采用酶联免疫吸附法检测上述细胞上清液中炎症因子TNF-α和IL-1β的浓度,评价黄芩配方颗粒和标准汤剂的抗炎活性。结果:10批次黄芩配方颗粒与黄芩标准汤剂中黄芩苷、汉黄芩苷和黄芩素的含量均无显著性差异,且对RAW264.7细胞无明显细胞毒性(50 mg·L-1)。与正常对照组相比,细菌脂多糖(LPS)显著诱导了巨噬细胞中NO,TNF-α及IL-1β(P<0.01)的分泌;与LPS组比较,50 mg·L-1的黄芩配方颗粒显著降低了细胞NO,TNF-α和IL-1β的水平(P<0.01),且与相同剂量的黄芩标准汤剂相比,抗炎效果没有显著性差异。结论:黄芩配方颗粒与黄芩标准汤剂的主要化学成分具有一致性,均可显著抑制LPS诱导的RAW264.7细胞释放NO,TNF-α和IL-1β,具有体外抗炎等效性,可为中药配方颗粒的研究和应用提供参考依据。 Objective: This study aimed to evaluate the efficacy equivalence of Huangqin dispensing granule and Huangqin standard decoction by investigating their anti-inflammatory activities in vitro. Methods:Based on the characterization of baicalin,wogonoside,baicalein content in Huangqin dispensing granule and Huangqin standard decoction by HPLC,the inflammatory model was established by stimulating mouse macrophage RAW264. 7 cells with LPS. The cytotoxicity and the effect on cell NO production of 10 batches of Huangqin dispensing granule and Huangqin standard decoction were determined by WST-1 and Griess method respectively.Then,the concentration expression of TNF-α and IL-1β in cell supernatant was detected by ELISA to evaluate theiranti-inflammatory activities. Results: There was no significant difference in the content of baicalin,wogonoside,baicalein in 10 batches of Huangqin dispensing granule and Huangqin standard decoction,which also exhibited no obvious cytotoxicity to RAW264. 7 cells at 50 mg·L-1. Compared with the normal control group,LPS significantly induced the secretion of NO,TNF-α and IL-1β( P 0. 01) in macrophages. Compared with LPS group,Huangqin dispensing granule at 50 mg·L-1 significantly decreased the levels of NO,TNF-α and IL-1β( P 0. 01),there is no significant difference compared with Huangqin standard decoction. Conclusion: There was no significant difference between the main chemical content of Huangqin dispensing granule and that of Huangqin standard decoction,which both significantly inhibited the release of NO,TNF-α and IL-1β in LPS-stimulated RAW264. 7 cells. It indicated that Huangqin dispensing granule and Huangqin standard decoction had the equivalent anti-inflammatory efficacy in vitro,which provided a reference for the application of TCM dispensing granule.
