小凹蛋白-1和基质金属蛋白酶-2在食管鳞癌中的表达及临床意义

Expressions of Caveolin-1 and Matrix metalloprotein -2 in esophageal squamous cell carcinoma tissues and their clinical significance

目的观察食管鳞癌(ESCC)组织中小凹蛋白-1(Caveolin-1)、基质金属蛋白酶2(MMP-2)的表达变化并探讨其意义。方法食管鳞癌组织110例(试验组),正常食管组织15例(对照组),用免疫组化法、反相-PCR法和免疫印迹法检测2组食管组织的Caveolin-1、MMP-2的表达,分析Caveolin-1、MMP-2阳性表达与ESCC临床病理参数的关系。结果试验组和对照组的Caveolin-1阳性表达率分别为73.64%(81例/110例)、33.33%(5例/15例);这2组的MMP-2阳性表达率分别为84.55%(93例/110例)、26.67%(4例/15例),试验组与对照组比较,差异均有统计学意义(均P<0.01);Caveolin-1和MMP-2在食管鳞癌中的表达呈正相关(r=0.430,P<0.0001);Caveolin-1、MMP-2阳性表达均与ESCC分化程度、TNM分期、淋巴结转移及肝转移相关(均P<0.05)。试验组及对照组Caveolin-1 mRNA的相对表达量分别为0.96±0.05,0.21±0.06;这2组的MMP-2 mRNA的相对表达量分别为0.89±0.03,0.23±0.05,试验组与对照组比较,差异均有统计学意义(均P<0.01)。结论食管鳞癌组织中Caveolin-1、MMP-2表达升高,其高表达与食管鳞癌分化程度、TNM分期、淋巴结转移及肝转移相关。

Objective To explore the expressions of Caveolin-1 and matrix metalloprotein-2（ MMP-2） in esophageal squamous cell carcinoma（ ESCC） tissues and discuss the clinical significance. Methods The expressions of Caveolin-1 and MMP-2 in 110 specimens of ESCC tissues（ treatment group） and 15 specimens of normal esophageal tissues（ control group） selected were detected by Immunohistochemistry technique,Reverse transcription-polymerase chain reaction（ RT-PCR）,and Western blot. The correlation between their positive expressions and the elinieopathologie features were analyzed. Results The positive rates of Caveolin-1 in treatment group and control group were 73. 64%（ 81 cases/110 cases） and 33. 33%（ 5 cases/15 cases）; the positive rates of MMP-2 in the 2 groups were 84. 55%（ 93 cases/110 cases） and26. 67%（ 4 cases/15 cases）,there were significant differences（ all P〈0. 01）. Expression of Caveolin-1 was positively related with the expression of MMP-2 in esophageal squamous cell carcinoma tissues（ r = 0. 430,P〈0. 0001）. The positive expressions of Caveolin-1 and MMP-2 were correlated with diffrerentiation degree,TNM stage,lymph node metastasis,and liver metastasis（ all P〈0. 05）. The relative expressions of Caveolin-1 mRNA in treatment and control groups were 0. 96 ± 0. 05 and0. 21 ± 0. 06; the relative expressions of MMP-2 mRNA in the 2 groups were 0. 89 ± 0. 03 and 0. 23 ± 0. 05,there were significant differences（ all P〈0. 01）. Conclusion The expression levels of Caveolin-1 and MMP-2 are high in ESCC tissues,which have correlation with diffrerentiation degree,TNM stage,lymph node metastasis,and liver metastasis.
