两种抑菌剂在铁皮石斛开放式组织培养的应用

Application of Two Bacteriostatic Agents in Open Tissue Culture of Dendrobium officinale

为降低铁皮石斛工厂化育苗的成本,简化组培程序,提高经济效益,探究铁皮石斛开放式组织培养的适用抑菌剂配方。以铁皮石斛为试材,在培养基中分别加入不同浓度的次氯酸钠(0%~0.2%)和代森锰锌(0%~0.03%)抑菌剂,以传统组培方法为对照,探究在铁皮石斛开放式组培中2种抑菌剂的最适使用浓度。结果表明:次氯酸钠和代森锰锌均可作为铁皮石斛开放式植物组织培养的抑菌剂,次氯酸钠浓度在0.025%以上,代森锰锌浓度在0.006%以上时,能最有效抑制培养基本身菌类及自然环境对培养基造成的污染。次氯酸钠浓度在0.2%以上,代森锰锌浓度0.024%以上能完全抑制人为接入菌类。当次氯酸钠浓度为0.15%时铁皮石斛不定芽增殖系数最高,达到2.48,代森锰锌浓度为0.020%时增殖系数最高,为2.68,均与对照无明显差异,抑菌效果理想,因此2种抑菌剂在铁皮石斛开放式组培上均具有良好的应用价值。

In order to reduce the cost of industrialized breeding of Dendrobium officinale,simplify the process of tissue culture and increase economic efficiency,the suitable antibacterial agent formula for open tissue culture of Dendrobium candidum was explored.In this study,Dendrobium candidum was used as the test material,different concentrations of sodium hypochlorite（0%-0.2%）and mancozeb（0%-0.03%）as bacteriostatic agent were added to the culture medium,traditional tissue culture as a control,the optimal concentration of two antimicrobial agents in open tissue culture of Dendrobium candidum was explored.The results showed that sodium hypochlorite and mancozeb could be used as antibacterial agents in open tissue culture of Dendrobium officinale.It demonstrated that the effective concentration of sodium hypochlorite was above 0.025%,the effective concentration of mancozeb was above 0.006% on reducing the pollution of the culture medium and the culture environment.When the concentration of sodium hypochlorite was above 0.200% and mancozeb concentration above 0.024% could completely inhibit the pollution caused by innoculated bacteria and fungi.The results showed that the multiplication coefficient of Dendrobium officinalein the open tissue culture were 2.48 at the concentration of 0.150% sodium hypochlorite and the multiplication coefficient were 2.68 at the concentration of 0.020% mancozeb.There was no significant difference between the two test groups and control group.Therefore,the two kinds of bacteriostatic agents had good application value in the open tissue culture of Dendrobium officinale.
