加味五味石膏汤对卵清蛋白致小鼠变应性鼻炎的作用

Effect of Modified Wuwei Shigao Decoction on OVA induced allergic rhinitis in Balb/c mice

目的:考察加味五味石膏汤对卵清蛋白(Optical Variable Attenuator,OVA)所致变应性鼻炎(Allergic rhinitis,AR)小鼠的治疗作用。方法:40只BALB/c雌性小鼠随机分为5组:空白对照组、OVA模型组、氯雷他定0.01 g/kg组、加味五味石膏汤5.46 g/kg、10.92 g/kg组。每组8只。除空白对照组外,其余均使用OVA制备AR模型。通过观察小鼠搔鼻和打喷嚏的行为,评估小鼠鼻部症状;采用HE及天狼星染色法观察鼻黏膜的炎症情况;采用ELISA法检测血清中免疫球蛋白E(Ig E)、肿瘤坏死因子(TNF)α、白细胞介素(IL)1β、IL-4和干扰素(IFN)γ的水平;采用Western Blot法检测鼻黏膜组织中转录因子T-bet和GATA-3蛋白的表达情况。结果:与空白对照组相比,AR模型小鼠的鼻部过敏症状较为明显,嗜酸性粒细胞的浸润显著,血清中Ig E、TNF-α、IL-1β、IL-4水平显著升高,IFN-γ水平显著性降低,而加味五味石膏汤5.46 g/kg、10.92 g/kg组能不同程度地显著改善小鼠的鼻部过敏症状,抑制血清中Ig E、TNF-α、IL-1β、IL-4的水平,增加血清中IFN-γ水平,增加鼻黏膜组织中T-bet的蛋白表达,降低GATA-3蛋白的表达量。结论:加味五味石膏汤对OVA所致小鼠AR具有治疗作用,与阳性药的治疗效果相当,其机制可能与调节Th1/Th2免疫失衡有关。

Objective： To investigate the therapeutic effect of Modified Wuwei Shigao Decoction on mice with Allergic rhinitis（ AR） caused by optical variable attenuator（ OVA）. Methods： Forty BALB/c female mice were randomly divided into 5 groups： control group,OVA model group,0. 01 g loratadine group,5. 46 g Modified Wuwei Shigao Decoction group and 10. 92 g Modified Wuwei Shigao Decoction group,8 in each group. OVA was used to establish AR model in all groups except for the control group. Nasal symptoms of mice were evaluated by observing the behavior of the murine nose and sneezing. Inflammation of the nasal mucosa was observed by HE and Sirius staining. Serum immunoglobulin E（ Ig E）,tumor necrosis factor（ TNF-α）,IL-1β,IL-4 and IFN-γ were detected by ELISA. Western blot was used to detect expressions of T-bet and GATA-3 in nasal mucosa. Results： Compared with the control group,allergic rhinitis symptoms in AR model mice were obvious,and the infiltration of eosinophils was significant. Levels of Ig E,TNF-α,IL-1β and IL-4 in serum were significantly increased.However,the level of IFN-γ was significantlu decreased. 5. 46 g and 10. 92 g groups of five flavour gypsum decoction significantly improved nasal allergic symptoms in mice（ P 〈 0. 01）,levels of serum Ig E,TNF-α,IL-1β and IL-4 were inhibited,the level of IFN-gamma in serum increased（ P 〈 0. 01）,the expression of T-bet protein in nasal mucosa and the expression of GATA-3 protein（ P 〈 0. 01） were increased.Conclusions： Modified Wuwei Shigao Decoction has the therapeutic effect on the mice with OVA-caused AR,which is equivalent to the treatment of positive drug. The mechanism may be related to the regulation of Th1/Th2 immune imbalance.
