摘要
目的:探讨三子养亲汤对支气管哮喘模型大鼠Th1/Th2平衡失调的作用机制。方法:将60只SD大鼠随机分成6组,每组各10只,分别为正常对照组、哮喘模型组、地塞米松0.75mg组、三子养亲汤1.5、3、6g/kg组。采用卵清白蛋白(OVA)与氢氧化铝致敏,并OVA雾化吸入激发制备大鼠支气管哮喘模型。各组大鼠经末次激发24h后,收集支气管肺泡灌洗液(BALF)及肺组织标本。采用酶联免疫吸附法(ELISA)检测大鼠BALF中的IFN-γ、IL-2和IL-5的含量;免疫组化法检测肺组织中T-bet、GATA-3蛋白的表达。结果:三子养亲汤6 g/kg组和地塞米松组可显著降低哮喘大鼠BALF中IL-5含量;地塞米松组和三子养亲汤3 g/kg、6g/kg组显著升高IL-2含量;三子养亲汤1.5g/kg、3 g/kg、6 g/kg组和地塞米松组显著升高IFN-γ含量。与哮喘模型组比较,三子养亲汤1.5g/kg、3 g/kg、6 g/kg组和地塞米松组可显著降低哮喘大鼠BALF中GATA-3含量;三子养亲汤3 g/kg、6 g/kg组显著升高Tbet含量。结论:三子养亲汤能够发挥抗支气管哮喘的作用,其机制可能与调控Th1/Th2平衡失调有关。
Objective: To observe the mechanism of Sanzi Yangqin decoction on Th1/Th2 imbalance in bronchial asthma. Methods: Sixty SpragueDawley rats were randomly divided into 6 groups,10 rats in each group: the normal group,asthma model group,dexamethasone group and Sanzi Yangqin decoction( 1. 5 g/kg,3 g/kg,6 g/kg). Rats were sensitized with ovalbumin( ova) and aluminum hydroxide( ova) and stimulated by OVA atomization inhalation to establish asthma model. Rats in each group were exposed to ovalbumin for 24 hours after the last stimulation. Bronchoalveolar lavage fluid( BALF) and lung tissues were collected. BALF and lung tissues were collected. Levels of IFN-γ,IL-2 and IL-5 in alveolar lavage fluid were detected by immunohistochemistry. Expressions of T-bet and GATA-3 in lung tissues were detected by immunohistochemistry. Results: The content of IL-5 in BALF of asthmatic rats was decreased in 6 g/kg Sanzi Yangqin decoction and dexamethasone groups. Contents of IL-2 increased in 3 g/kg and 6 g/kg groups. Contents of IFN-γ increased in 1. 5 g/kg、3 g/kg、6 g/kg Sanzi Yangqin decoction groups and dexamethasone group. Compared with the asthmatic model group,GATA-3 protein contents in lung tissues of asthmatic rats were significantly decreased in the 3 g/kg and 6 g/kg groups. Contents of T-bet protein were increased in 3 g/kg and6 g/kg groups,and differences were statistically significant( P 〈 0. 05). Conclusion: Sanzi Yangqin decoction can play the role of antiasthma,and its mechanism may be related to regulating the imbalance of Th1/Th2 balance.
