摘要
目的:为探讨桑皮素对Du145细胞的增殖抑制及其诱导凋亡的影响。方法:用5、10和15μmol/L的桑皮素作用于Du145细胞24 h后,在倒置显微镜及透射电镜下观察细胞形态学变化;利用CCK-8法来检测桑皮素对Du145细胞的增殖的影响;应用流式细胞术和DNA fragmentation实验检测Du145细胞的凋亡情况;采用Western blot技术检测Gli1和VEGF蛋白的表达变化。结果:发现桑皮素能够抑制Du145细胞的生长,并诱导Du145细胞发生凋亡甚至死亡,作用24h后IC50=16.5μmol/L;与对照组相比,细胞凋亡率显著升高,当作用浓度≥15μmol/L后细胞晚期凋亡明显增多;随着桑皮素作用浓度的增加,Gli1和VEGF蛋白表达量明显下降。结论:桑皮素诱导Du145细胞的凋亡,并降低了Hh信号通路中Gli1和VEGF因子的表达。
Objective: To investigate the proliferation inhibition and inducing apoptosis of mulberin on Du145 cells. Methods: Du145 cells were treated with 5,10,15μmol/L mulberin for 24 hours,morphological changes of cells were observed under inverted microscope and transmission electron microscopy. The inhibitory effect of mulberin on Du145 cells was detected by CCK-8 method. The apoptosis of Du145 cells was detected by flow cytometry and DNA fragmentation. Expressions of Gli1 and VEGF protein were detected with Western blot. Results: Mulberin inhibited the growth of Du145 cells,and it induced the apoptosis even death of Du145 cells( when IC50= 16. 5 μmol/L after 24 hours). Compared with the control group,the apoptosis rate of Du145 cells was significantly increased. And the late apoptosis obvious increased when the concentration was ≥ 15 μmol/L. With the increase of the concentration of mulberin,protein expressions of Gli1 and VEGF significantly decreased. Conclusion: Mulberin can induce the apoptosis of Du145 cells,and it can decrease expressions of Gli1 and VEGF factors in the Hh signaling pathway.
