破壁、未破壁雨生红球藻粉体外抗氧化活性及体内抗衰老作用及比较研究 被引量:9

Study on and comparison antioxidant activities in vitro and anti-aging effects in vivo between Haematococcus pluvialis powders and broken Haematococcus pluvialis powders

摘要 目的:探究破壁、未破壁雨生红球藻粉的体外抗氧化活性及体内抗衰老作用,量化并比较两者活性及作用差异,为应用雨生红球藻提供选择依据。方法:以ABTS、DPPH及羟自由基清除率及IC50值为指标,评价破壁、未破壁雨生红球藻粉的体外抗氧化活性。采用2月龄SD大鼠作为青年正常对照,12月龄SD大鼠作为中老年模型对照,灌胃给予破壁、未破壁雨生红球藻粉(90mg/kg),7周后测定大鼠血清及脑组织抗氧化指标、皮肤相关指标、血脂水平及肝功能指标。结果:体外抗氧化实验结果显示,破壁、未破壁雨生红球藻粉对ABTS、DPPH及羟自由基的清除率随浓度增加而升高,而在总计88.9%的浓度检测点,破壁雨生红球藻粉的体外自由基清除率高于未破壁雨生红球藻,破壁雨生红球藻粉的体外自由基清除率IC50值更低。体内抗氧化指标显示,破壁、未破壁雨生红球藻粉均能升高中老年大鼠血清T-OAC水平、GSH-Px及CAT活力,而破壁雨生红球藻粉还能降低血清及脑组织MDA含量,升高血清GSH、脑组织GSH水平及SOD活力;皮肤相关指标显示破壁、未破壁雨生红球藻粉均能降低皮肤组织Tyr活性,而破壁雨生红球藻粉还能升高Hyp水平;血脂水平指标显示破壁雨生红球藻粉能降低血清TG含量;肝功能指标显示未破壁雨生红球藻粉能降低血清AST活性。结论:破壁、未破壁雨生红球藻粉均具有较好的体外抗氧化活性及体内抗衰老作用;破壁雨生红球藻粉具有潜在的降血脂功效,未破壁雨生红球藻粉具有潜在的肝保护作用;破壁雨生红球藻粉较未破壁的体外抗氧化活性及体内抗衰老作用更显著、全面,结合市场价格在两者中显示出更优的性价比,为应用雨生红球藻提供了选择依据,值得进一步研究与开发。 Objective,: To investigate antioxidant activities in vitro and anti-aging effects in vivo of Haematococcus pluvialis powders and broken Haematococcus pluvialis powders,quantify and compare differences between them,and provide a evidence for the usage of Haematococcus pluvialis. Methods: ABTS,DPPH and hydroxyl radicals scavenging rates and their IC50 were used as indexes to evaluate antioxidant activities in vitro of Haematococcus pluvialis powders and broken Haematococcus pluvialis powders. 2-month-old SD rats as the youth control,12-month-old SD rats as the aged model,Haematococcus pluvialis powders and broken Haematococcus pluvialis powders( 90 mg/kg) were administered. After 7 weeks,serum and brain tissue antioxidant index,skin related indicators,blood lipid levels and liver function indicators were measured. Results: Clearance rates of ABTS,DPPH and hydroxyl radicals increased with the increase of concentrations of Haematococcus pluvialis powders and broken Haematococcus pluvialis powders. However,in a total of 88. 9 % of concentration detection points,the free radical scavenging rate of broken Haematococcus pluvialis powders was higher than that of unbroken. Haematococcus pluvialis powders had lower IC50 value. Antioxidant indexes in vivo showed that the broken and unbroken Haematococcus pluvialis powder raised serum T-OAC,GSH-Px and CAT in aged rats,but broken Haematococcus pluvialis powders reduced serum and brain MDA,increased serum GSH,brain tissue GSH and SOD. Skin related indicators showed that broken and unbroken Haematococcus pluvialis powder reduced Tyr in skin tissues,while broken Haematococcus pluvialis powder also increased the skin Hyp. Lipid level indicators showed that the broken Haematococcus pluvialis powders reduced serum TG. Liver function indicators showed unbroken Haematococcus pluvialis powders reduced serum AST. Conclusion: Haematococcus pluvialis powders and broken Haematococcus pluvialis powders both show good antioxidant activities in vitro and antiaging effects in vivo. Haematococcus pluvialis powders has the potential hypolipidemic effect,and unbroken Haematococcus pluvialis powders has the potential liver protection. However,the antioxidant activity in vitro and anti-aging efficacy in vivo of broken Haematococcus pluvialis powders were more significant and comprehensive,which show the higher price/performance ratio. These results provide a evidence for choice.
