复方当归妇炎微灌肠剂的抗炎作用及机制研究 被引量:6

摘要 目的:研究复方当归妇炎微灌肠剂的抗炎作用及作用机制。方法:建立二甲苯致小鼠耳肿胀、角叉菜胶致大鼠足肿胀炎症模型,观察复方当归妇炎微灌肠剂的抗炎作用;建立苯酚胶浆致大鼠盆腔炎模型,造模8天后,各组直肠及灌胃给药14天,以ELISA法测定大鼠子宫、输卵管、卵巢组织肿瘤坏死因子α(TNF-а)、白细胞介素-2(IL-2)、白细胞介素-6(IL-6)、前列腺素E2(PGE2)的含量;用免疫组化法检测NF-κB p65、Caspase-3在子宫、卵巢组织中的表达。结果:复方当归妇炎微灌肠可以明显抑制二甲苯致小鼠耳廓肿胀,减轻角叉菜胶致大鼠足跖肿胀;与模型对照组比较,复方当归妇炎微灌肠剂17.5g/kg组、8.75g/kg组和妇炎康组能够改善大鼠子宫、卵巢、输卵管的病变程度,降低大鼠血清、子宫、卵巢中IL-2、IL-6、TNF-α、PGE2的含量;复方当归妇炎微灌肠剂17.5g/kg组、8.75g/kg组和妇炎康组均能显著抑制NF-κB p65在子宫、卵巢的阳性表达;复方当归妇炎微灌肠剂的各个剂量均能显著提高Caspase-3在子宫、卵巢的阳性表达。结论:复方当归妇炎微灌肠剂对炎症具有良好的抑制作用。复方当归妇炎微灌肠剂治疗苯酚胶浆致大鼠慢性盆腔炎的作用机制可能是通过抑制促炎因子的表达,促进炎细胞调亡等方式,改善盆腔炎子宫的肿胀和粘连。 Objective: To study the anti-inflammatory action and mechanism of Compound Angelica Fuyan Micro-Enema. Methods: The mice ear edema induced by xylene and the rat foot swelling induced by carrageenan were used to research the anti-inflammatory action of Compound Angelica Fuyan Micro-Enema. Rats with pelvic inflammatory disease were induced by the phenol glue. 7 days after modeling,rectal administration and gavage administration were performed in each group for 14 days. The amount of TNF-α,IL-2,IL-6 and PEG2 were determined by ELISA. Expressions of NF-κBp65,Caspase-3 in uterus and ovarian tissues. Results: Compound Angelica Fuyan Micro-Enema obviously inhibited mice ear edema induced by xylene,and relieved the rat foot swelling induced by carrageenan. Compared with the model group,the high( 17. 5 g/kg),middle( 8. 75 g/kg) doses of Compound Angelica Fuyan Micro-Enema and Fuyankang obviously improved lesions extents of rat uterus,ovaries and fallopian,and decreased contents of IL-2,IL-6,TNF-α,PGE2 of rat serum,uterus and ovary. The high( 17. 5 g/kg),middle( 8. 75 g/kg) doses of Compound Angelica Fuyan Micro-Enema and Fuyankang inhibited expressions of NF-κBp65 in uterus and ovary. Various doses of Compound Angelica Fuyan Micro-Enema significantly improved expressions of Caspase-3 of uterus and ovary. Conclusion: Compound Angelica Fuyan Micro-Enema has a significantly anti-inflammatory action. In the treatment of rats with chronic pelvic inflammatory disease induced by the phenol glue,the mechanism of Compound Angelica Fuyan Micro-Enema to improve the swelling of the uterus and adhesion caused by pelvic inflammatory may be related to inhibiting the expression of cytokines or promoting inflammatory cell apoptosis.
